There were much nutrients in vinasse from sugarcane molasses alcohol,and it could be used as the medium of microorganisms.Humic acid contain a variety of active functional groups,and now it is used widely.In order to make use of vinasse to product humic acid,This paper has made the following research.First of all,The H812strain was screened from sludge through appropriate screening model,and it could make use of vinasse to product humic acid.The H812strain was preliminarily identified as Aspergillus sp by its culture characteristics,morphological features and ITS sequence analysis.Subsequently,The concentration of vinasse and strain culture conditions were optimized by the monfactorial researchs and orthogonal experiments.The results showed that16°Bx was the optimal concentration of vinasse.The optimal fermentation conditions were:8days,34℃,200r/min,pH7.0,inoculum size12%, liquid volume50mL/250mL.Applicated optimized fermentation conditions, HA content fermented with H812strain was38.12g/L,which is as1.48times than before.After fermented by H812strain,the COD, BOD,total sugar,Brix,total nitrogen of vinasse were decreased by87.12%,70.36%,60.31%,59.03%and 70.36%respectively.Finally,Extration process of biotechnology humic acid was optimized by monfactorial experiments and orthogonal tests.The results showed that the optimal process was as follows:the biotechnology humic acid was extract with the mixture of6mol/L NaOH and0.025mol/L Na4P2O7solution for three hours in26℃,the extraction amount was35%,then acided to pH1.5with HCl.The biotechnology humic acid was characterized by ultraviolet-visible absorption spectrometry,Infrared Spectroscopy,fluorescence spectrum and chemical methods.The results indicated that BHA has many characteristics,such as Low aromatic degree,less unsaturated bond,higher pH value,higher flocculation limit and so on.
