Recently, capillary electrophoresis (CE) has been utilized for theseparation of all kinds of analytes because of its advantages of highefficiency, rapidity, and small sampling volume. Along with thedevelopment of CE, various computer simulation technologies of CEhave been proposed, among which HVL function is the important methodof simulation of CE peak shape. Free flow electrophoresis (FFE) issuitable for separation and purification of biological materials, with itsadvantages of continuous separation, mild separation conditions, andclearance of solid support media. Due to many factors which affectseparation efficiency of FFE system, the existing mathematical modelsare too complicated.In this paper, a simplified mathematical model of capillary zoneelectrophoresis was built by using the modified-HVL function. Thismodel just considers mobility, concentration and pKa values of matter.And computer simulation software of the above model were designed and implemented in Borland Delphi7environment. It can calculate the pH,ionic strength, conductivity and resistance of BGE and also calculate theeffective mobility, diffusion coefficient and ion concentration of analyteions according to the material parameters and system parameters input byusers. Then the dynamic separation process was output using somealgorithm. With DMSO and histamine as analytes, we can get the actualelectropherogram. By the comparison of the electropherogram betweenexperiment and computer simulation, reliability and veracity of thesoftware were validated.20amino acids were separated using thesoftware, and the comparisons demonstrate the high agreement betweenthe simulation results and the experimental results cited from references.As there is a certain relation between FFE and CE, the mathematicalmodel of CZE was conversed to the model of FFE, in which the equationof band-broadening was simplified to avoid being too complex. Thenaccording to the model described above the simulation software of FFZEwas compiled, which can also calculate some material parameters andsystem parameters, and display dynamic separation electropherogram ofFFZE. Finally, this software simulated separation process of20aminoacids, and the electropherogram was compared with that of CZEsimulation software; the results was satisfying, and the correctness ofFFZE simulation was verified.
