Natural medicines (NMs), such as Traditional Chinese medicines (TCMs), play animportant role in the treatment of diseases because of their reliable clinical performance. In thisthesis, HPLC and immobilized liposomes chromatography (ILC) were used to analyze thebiological activity and transmembrane permeability of components in Hippocampus, Cassiaobtusifolia L., etc. And the study provides a new technology and method for analyzing activeingredients of NMs.In this work, qualitative and quantitative analyses of the compounds in Gastrodia elata BI.and Cassia obtusifolia L. were developed by RP-HPLC coupled with UV-visible spectroscopy,respectively. Under Isocratic conditions, one bioactive compound in Gastrodia elata BI wasidentified as Gastrodin, under gradient conditions, four bioactive compounds in Cassiaobtusifolia L. were identified as Aloe-emodin, Emodin, Chrysophanol and Physcion. In addition,the concentrations of five bioactive compounds were determined.An ILC stationary phase were prepared and used to analyze the transmembranepermeability of components in the ethanol extract of Gastrodia elata BI. and Cassia obtusifoliaL.. The results showed that at least seven kinds of active ingredients in Cassia obtusifolia L. cansignificantly retained on an ILC column, and three of them were identified as aloe-emodin,Emodin and Chrysophanol. But Gastrodin from Gastrodia elata BI. did not show anysignificantly retention.The protein contents in the male and female Hippocampus were analyzed by Kjeldahmethod, respectively: protein content of male Hippocampus was55.83%, that of female one was49.00%for. And the ash content was also determined: the male Hippocampus was12.60%, thefemale Hippocampus was12.30%. A simple and sensitive method for anaIysis of the aminoacids compositions in Hippocampus were developed by pre-coIumn derivatization-HPLC withthe neutral protease digestion. The hydrolysis time was360min, neutral protease enzymeconcentration was1.000mg/mL. The results showed that17amino acids, including8essentialamino acids, detected in the enzymatic hydrolyzate of Hippocampus. The percentage of essentialamino acids content was more than60%of the total amino acids. At the same time, antioxidantactivity, antibacterial activity and antifatigue activity of the enzymatic hydrolyzate weremeasured with the pharmacological models. The results indicated obviously that the enzymatichydrolyzate of Hippocampus showed multiple pharmacological activities.
