Molecular imprinting technology is a method for preparing a particular molecule,which has a specific recognition performance polymer. It shows a broad prospect fordevelopment in the separation and purification, immunoassays, biosensors and otherfields.Preparing principles and methods of molecular imprinting technology and itsapplication were reviewed in detail, at the same time, some current problems and trendswere discussed as well in this paper. Experiment mainly includes three parts: in the firstpart, solvent and functional monomers involved in the pre-polymerization process wasscreened and the interaction between template molecules and functional monomer atdifferent concentrations was assessed. In the second part, amination silica gel was usedas carrier and surface molecular imprinting method was applied to optimize the threefactors in the polymer preparation process. imprinted polymer was prepared in anacetonitrile aqueous system presenting a specific adsorption of glucose polymers byhydrogen bonding. Then tatic adsorption capacity and competitive adsorption was usedfor measuring its characteristics. Scanning electron microscopy, elemental analysis, IRspectroscopy and other techniques were used to analyze and characterize the imprintedpolymers. In the third part, molecularly imprinted polymers were made into a solidphase extraction column, then the adsorption and elution conditions were optimized.Results showed that: firstly, there are some factors, such as the electrostatic effectbetween template molecule and functional monomers in the polarity condition, thetemperature, time and proportion of pre-polymerization, having significant impact onthe effect of the pre-polymerization. The optimum conditions are as followed:temperature is20-30℃, prepolymerization time is2h and the function modulesmolecular ratio is1:4. Secondly, molecularly imprinted polymer which was preparedwith100mg crosslinking agent,20mg initiator and1.0g silica has the largest adsorptioncapacity. the dosage of crosslinking agent and silica have a greater impact on thepolymer property. Thirdly, a solid phase extraction column prepared using0.5mgpolymer was loaded with5.0ml glucose sample (2.0mg/ml) eluted in an acetonitrileaqueous(4:1) system,which has more than90%adsorption amount. more than90percent recovery can be obtained when the water, different ratios of methanol and aceticacid was used as the eluent.
