Many efforts have focused on identification and pharmacological research of activeingredients of traditional Chinese medicines (TCMs). In this paper, microultrafiltration-HPLCanalysis method was developed and carried on the interaction and qualitative analysis for theactive components from Stephania tetrandra, Artemisiacapillaris Thunb, salvia miltiorrhizadropping pill.In this work, we firstly estabolished the methods of qualitative and quantitative analysis fortwo main effective components in the ethanol extract of Stephania tetrandra, ArtemisiacapillarisThunb, salvia miltiorrhiza dropping pill using HPLC coupled with UV-Vis spectrum.7activecompounds from the aboved three extracts were identified, and the concentrations of them inmedicinal materials were also determined.We adopted the microultrafiltration-HPLC analysis method to investigate activities of thebinding for the active ingredients from Stephania tetrandra, Artemisiacapillaris Thunb andsalvia miltiorrhiza dropping pill with ct-DNA, mtDNA and rat hepatic DNA. The results showedthat the binding activity of Tetrandrine and FangchinoLine with DNA is higher than that ofactive compounds, SaLvianicacid A sodium, protocatechuic aldehyde from salvia miltiorrhizadropping pill show lower binding activities with ct-DNA. At the same time, the binding types ofactive compounds had been studied by means of UV-Vis spectrum, the spectra indicated that thebinding type of Tetrandrine and FangchinoLine with ct-DNA, mtDNA is embedded type, and thebinding type of FangchinoLine with ct-DNA, mtDNA is groove type.For further investigating binding properties of active ingredient with DNA, the compectiveproperties of active compounds interacted with mtDNA was investigated. The results indiactedthat the binding degree of FangchinoLine increased when the concentration of Tetrandrine is lessthan125μg/mL. But the concentration of Tetrandrine (competition reagent) continues to increase,the binding degree of both of them with mtDNA decreased. On the other hand, when theconcentration of Tetrandrine is125μg/mL, the concentration of FangchinoLine graduallyincreased from50μg/mL, the binding degree of FangchinoLine and Tetrandrine with mtDNA didnot show obvious changes. It can be concluded that binding activity of Tetrandrine with mtDNAis higher than that of FangchinoLine. In the same way, the binding degree of Scoparone andhydroxy acetophenone was observed increased with increasing the concentration of hydroxyacetophenone from50μg/mL to100μg/mL. It indicated that it is synergistic action. When theconcentration of the competitive reagent continued to increase, the binding degree of them withmtDNA decreased and indicated that it is competitive action.
