Polyphenols was the main natural active constituents in the Castanea mollissima Blume involucres, which has a high value in food and medicine industries. In this study, the main components of Castanea mollissima Blume involucres polyphenols extraction, purification and its antioxidant activities were evaluated. High-performance liquid chromatography-mass spectrometry was used to determine the main components of of Castanea mollissima Blume involucres polyphenols. The results are as follow:1, By optimization of single factor and response surface experiment, ultrasonic technology was applied to extract polyphenol, the optimal technological conditions were as follows:the ethanol concentration40%, liquid ratio32:1, ultrasonic temperature67℃, working time38min. The corresponding extraction yield of polyphenol from Castanea mollissima Blume was23.20%, with extraction rate47.86%.2, The orthogonal test was adopted and the factors influencing the process parameters were determined, water technology was applied to extract polyphenol. The results showed that optimal conditions for the extraction:ethanol concentration was30%, liquid-solid ratio was18:1(mL:g), extraction temperature was80℃, extraction time was140min. The corresponding extraction yield was22.61%, with of polyphenol concentration of51.23%.3, With the aim to purify polyphenols from Castanea mollissima Blume by macroporous resin, macroporous resins were tested for their static adsorption/desorption performance towards polyphenols. The results showed that AB-8was the optimal macroporous resin for purifying polyphenols, the optimum purifying condition was as follows: the mass concentration and the sampling rate were lmg/mL and2mL/min, respectively. polyphenol sample volume was400mL.Whereas the optimal desorption conditions are found to be a flow rate of1mL/min using140mL of70%ethanol for elution. The purity of polyphenols from Castanea mollissima Blume increases from47.86%to81.67%with ultrasonic technology, the purity of polyphenols from Castanea mollissima Blume increases from51.23%to86.54%with water technology, respectively.4, The antioxidant activities were evaluated through several biochemical assays(DPPH、ABTS and POV) and in vitro cell experiment(cell proliferation; T-AOC,T-SOD and GSH-Px activities; MDA, and GSH content). The results showed that polyphenols from Castanea mollissima Blume increases had strong antioxidant activities and it seemed to have positive effects in mitigating heat stress.5, HPLC-MS was used to determine the main components of of Castanea mollissima Blume involucres polyphenols. The results showed that the main component of CBI was ellagic acid (18.64mg/g).
