Russula polysaccharide is a fungus that has high nutritional value and high exploitationpotential. In this dissertation, we studied in vitro antioxidant activity and inhibition ofproliferation of Hela cells and other properties using Russula polysaccharide fruiting bodiesas raw materials. The main contents and conclusions were as follows:We adopted three different methods: the traditional red mushroom polysaccharide hotwater extraction, ultrasonic assisted extraction, microwave assisted extraction ofpolysaccharides. Therefore, we selected extraction time, Russula polysaccharide liquid ratioand extraction temperature, microwave power as a research object, then we researchedevaluation of red mushroom polysaccharide extraction rate. To choose the optimal extractionyield and condition, we compared comprehensively of the three methods using single factortest and found that microwave-assisted extraction method could get the higher extraction ratein a short period of time. So we finally selected microwave-assisted extraction method to gainthe optimal extraction condition by conducting three factors and three levels Box-Menkencentral composite experiment with the response surface. The optimal extraction conditionswere as follows: Microwave time5.9393min, microwave power498.909W, liquid-solid ratioof50(ml/g), in such condition, actually the extraction rate was7.288%, almost same as thetheory whose extraction rate was7.517%.The traditional hot water extraction of polysaccharides, ultrasonic extraction andmicrowave assisted Russula polysaccharide can,to some extent, remove·OH radicals, freeradical O一2and NO一2, but the effect is far less than Vc.·OH radical scavenging activity isthe maximum among three methods, then the scavenging effects of O一2, and the nitritescavenging effect is the least.Russula polysaccharide is extracted by above three methods, its inhibition effect onHela cells was determined by MTT and cell morphology was also studied. The results showedthat treatment of Hela cells after48h with the three methods for the extraction of Russulapolysaccharide has obvious inhibitory effect on the growth of Hela cells, and increases withthe increase of the concentration of polysaccharide; in addition, with the concentrationincreasing, cell morphological characteristics similar to apoptosis. In the meantime, we cometo the conclusion that traditional hot water extraction polysaccharides is of the best effect ofthe three methods, later, microwave assisted extraction;effect of ultrasonic assisted extractionis poor. The specific inhibition mechanism remains to be studied further.China is rich in wild mushroom resources, but is short of development, mainly in fresh or dry products, low added value. According to the results of this study, Russula polysacchar ideeffects on good anti-oxidation and significantly inhibit Hela tumor cells, can be used as rawmaterials for functional beverages, health food and anti tumor drug, which has very broadprospects for development. At the same time, along with the in-depth study of Russulapolysaccharide anti-tumor effect, it will bring the gospel to the majority of cancer patients,and make great contribution to the cause of human health.
