The plant polysaccharides have low toxicity and a amount of biological activity,which become a research hotspot in the field of medicine.Loranthaceae is used as a clinical common traditional Chinese medicine,which has been listed as a top grade by Shennong's Classic of Materia Medica,the polysaccharides of loranthaceae currently is focused on the Scurrula parasitica L.Mistletoe of camellia oleifera(M)parasitized on the oil tea,which is belongs to Helixanthera Lour.,the polysaccharides from M has been very few studied in any depth.Therefore,the purpose of this study is to explore the optimum extraction technology of polysaccharides,analyze the structural characteristics and biological activities of the extracted polysaccharides.Research on the M polysaccharides provides reference of the research on comprehensive development and utilization of M.The single factor and response surface methodology(RSM)were used to determine the optimum extraction process for polysaccharides from the branch of M by water as solvent.The nine kind of crude polysaccharides were extracted from the three parts of M(roots,branches and leaves)by different solvents(hydrosolvent,acid solvent,alkali solvent)and alcohol precipitation.The method of phenol sulfuric acid method,BCA method and meta-hydroxybiphenyl method were used to measure the total sugar contents,protein contents and uronic acid contents of nine polysaccharides from M.The main structural characteristics of polysaccharides from M were determined by IR,GC and GPC.The vitro antioxidant and anticoagulant activities of these polysaccharides were detected in antioxidation experiments(ABTS,DPPH,CAA)and anticoagulant experiments(APTT,PT,TT),which was analyzed the relationship with structure,extraction solvent and extraction sites.The conclusions are as follows:(1)The optimum extraction conditions were temperature of 95.55 ?,times of 2.59 times,time of 2.2 h,solid-liquid ratio of 10.09 mL/g,and the predicted maximum yield of polysaccharides was 2.67%.The optimum extraction conditions were adjusted when taking into account the actual extraction,as follows:temperature 95 ?,times 2 times,time 2 h,solid-liquid ratio 10 mL/g,and the predicted maximum yield of polysaccharides was 2.49%,the actual maximum yield of polysaccharides was 2.49±0.2%.(2)The total sugar content,uronic acid content,protein content,average molecular weight,and monosaccharide composition were affectted by the extraction sites and extraction solvents.(3)The molecular weight of alkaliextracted polysaccharide from M was the smallest in the same extraction site.(4)Polysaccharides from M were mainly consisted of Rha,Ara,Xyl,Glc,Man,and Gal,but the ratio of molar were different with each other.(5)Infrared spectrum analysis revealed that the polysaccharides of M were a configuration and ?configuration with pyranose.(6)The anticoagulant experiment displayed that the nine kind of polysaccharide extracted from M had anticoagulant activity in vitro.The uronic acid contents,galactose content and extraction site could influence the anticoagulation effect.(7)The antioxidant capacity of the M polysaccharides were affectted by the extraction solvent,extraction site and average molecular weight,anticoagulant activities were negative correlated with the average molecular weight.In this study,it hopes to provide scientific basis and technical support for the development a series of M polysaccharides.
