In the field of drug research, more than one-third of the clinical drug is from the nature of the natural products. Based on the structure of the active compounds of natural products in nature, Organic chemists are constantly exploring new ways to combine and restructure hydrocarbon molecules to make new Pharmaceuticals with higher activity and low toxicity, and make them widely used in clinical. This is one of the important ideas in the development of the new drugs. New structural, curative effect and low toxic compounds were got from natural products, which could be used for drug research. This dissertation was consisted of two parts:1. The separation, purification, structure conformation of chemical constituents of Brucea javanica (L) Merr.Brucea javanica (L) Merr. was obtained from Bozhou Guo Shao Tang Tech Co., Ltd. Company of Anhui, powdered and extracted with99%methanol for three times. Then the resulting mixture, which was obtained via vacuum distillation unit, was diluted with methanol, and extracted with equal volume of petroleum ether for three times. The resulting mixture, which was obtained via vacuum distillation from the methanol layer, was isolated by repeated column chromatography (CC) on silica gel, RP-18, and Sephadex LH-20, as well as by preparative thin layer chromatography (PTLC) and recrystallization. From Brucea javanica (L) Merr.,20compounds were isolated. The structures of these compounds were elucidated by means of spectroscopic methods including UV, EI-MS,1D-NMR (’H-NMR,13C-NMR) and2D-NMR (HMBC and HMQC), and12compounds were identified as follows by spectral analysis:Y-1:Bruceine DY-2:Bruceine EY-3:UracilY-4:p-Hydroxybenzoic acidY-5:p-StigmasterolY-6:p-Sitosterol Y-7:Stigmasterol-3-O-β-D-glucopyranosideY-8:New compoundY-9:AdenosineY-10:Yadanzioside BY-11:AllantoinY-12:Palmitic acidAmong them, Y-8was a new compound, Y-11was isolated from Brucea for the first time, and Y-3, Y-4, Y-7were isolated from this plant for the first time.Heptadecanoate methyl ester, oleate methyl ester, octadecenoic acide ethyl ester were found to be the main components of volatile oil from Brucea javanica by GC-MS.The antitumor acitivity in vitro of Bruceine D were evaluated by standard MTT assay. We selected three kinds of tumor cell lines:huaman lung cancer cells(A549), human colon cancer cells(HCT116) and human breast cancer cells(MD-MB-231). The results showed that Bruceine D exhibited the high activity on cancer cells HCT116and other tumor cells to some extent.2. Company of Anhui, powdered and extracted with95%methanol for three times. Then the resulting mixture, which was obtained via vacuum distillation unit, was diluted with water, and extracted with ethyl acetate and dichloromethane. The alkaloid constituents and non-alkaloid constituents were obtained from Stephania tetrandra S.Moore. The two sections were isolated by repeated column chromatography (CC) on silica gel, RP-18, and Sephadex LH-20, as well as by preparative thin layer chromatography (PTLC) and recrystallization. From Stephania tetrandra S.Moore,20compounds were isolated. The structures of these compounds were elucidated by means of spectroscopic methods including UV, EI-MS,1D-NMR (1H-NMR, C-NMR), and2D-NMR (HMBC and HMQC), and8compounds were identified by spectral analysis:N-1:TetrandrineN-2:FangchinolineN-3:CycleanorineN-4:N-Hydroxy DiethylamineN-5:Palmitic acidN-6:StigmasterolN-7:SitosterolN-8:New CompoundAmong them, N-8was a new compound.
