Broussonetia papyrifera and Arctium lappa are widely distributed in our country, havingan edge over relative natural resources, and the planting area is expanding year by year.Thenutrients of Broussonetia papyrifera leaves are very rich and to an analysis of its proteincontent as high as20%~30%. The vitamins, carbohydrates and trace elements are alsoabundance in Broussonetia papyrifera leaves.The same to the Arctium lappa leaves which arealso high values in nutrients.Its mainly rich in polysaccharide(mostly inulin), cellulose,protein, calcium, phosphorus, iron,variety of vitamins that human body needs and minerals.In this study, Broussonetia papyrifera leaves and Arctium lappa leaves are as the mainlymaterials.By the method of Alkali extraction, successfully extracted polysaccharide andprotein from two kinds of leaves at the same time. Then separate the two kinds of bioactivesubstance through the method of alcohol precipitation and the characteristic of isoelectricpoint. Used the method of Phenol-sulfuric acid and Kjeldahl to measure the content ofpolysaccharide and protein respectively.Simultaneous extraction of polysaccharide and protein from Broussonetia papyriferaleaves by Alkali extraction was studied. By single factor and orthogonal test the optimumconditions are as follows: the alkali concentration1%,extraction time5h, extractiontemperature70℃, the liquid-solid ratio35:1, pH3. By the above conditions, the extractionyield of polysaccharide is28.39%, and the extraction yield of protein59.59%.Simultaneous extraction of polysaccharide and protein from Arctium lappa leaves byAlkali extraction was studied. The optimum extraction conditions were confirmed by themono-factor and the orthogonal experiment.The optimum extracting conditions were: alkaliconcentration of2%,extraction time of5h, extraction temperature of80℃, ratioof liquid tosolid of25:1, pH5. By the above conditions, the extraction yield of polysaccharide was45.30%, and the extraction yield of protein was53.25%.The structure of the Broussonetia papyrifera leaves and Arctium lappa leaves was analyzed by IR. Through the contrast of leaves before reaction and after reaction can bepreliminary make an character on quantitative and qualitative.
