Fullerene is a kind of Carbon simple substance that the graphite evaporates, it has a variety of biological functions. Fullerenols is the mulit-hydroxy fullerene, which has allylic hydroxyl and middle electron affinity. Fullerenols can work as the antioxidant in organisms, it is easy to dissolve in water and to be utilized by these organisms. In order to understand the ultraviolet protective effect of fullerenes and fullerenols, taking the chicken embryo fibroblasts cells as the research objects, this thesis attempts establish the ultraviolet damage cell model and to explore the protective effect of fullerenes and fullerenols for ultraviolet damage cell model adopting the method of ultraviolet irradiation. The main findings were obtained as follows:(1) Multi-hydroxy fullerenols which was easy to dissolve in water at room temperature is prepared using organic synthesis method. And the multi-hydroxy fullerenols is shown adopting three methods:fourier transform infrared spectroscopy (FT-IR), Mass spectrum (MS), H nuclear magnetic resonance (’H-NMR) methods.(2) The ultraviolet damage cell model is established using ultraviolet irradiation method: the distance of UV irradiation is42cm, the wattage of UV irradiation is25W, the time of UV irradiation is7min.(3) This thesis analyzes the effect of fullerene and fullerenols for ultraviolet damage cell model, experimental results illustrate:different concentrations of fullerene can improve cell survival rate of ultraviolet damage cells model slightly. Different concentrations of fullerenols have the protective effect for ultraviolet damage cells model. MTT detection results improve that cell survival rate of the experimental group arrives at92precent, cell survival rate of the control group arrives at50.2precent. The optimal concentration of fullerenols for ultraviolet damage cells model is200μg/ml, the best adding time is2hours before irradiation. After the detection, for the experimental group, the intracellular MDA content was decreased from64±2.3nmol/106to15.5±2.6nmol/106and the extracellular MDA content was decreased from17.3±0.83nmol/106to5.4±0.87nmol/106. For both the intracellular and extracellular, the content of SOD and CAT were increased correspondingly. The results show that:fullerenols has the protective effect of ultraviolet damaged chicken embryo fibroblasts cells.
