Abstract:This paper examines a variety of chromatographic techniques, including high performance liquid chromatography-diode array detection (HPLC-DAD), gas chromatography-electron capture detector (GC-ECD) and gas chromatography-tandem mass spectrometry (GC-MS), for the determination of ferulic acid in a variety of traditional Chinese medicine and persistent organic pollutants in corals. The established high-performance liquid chromatography was successfully used for pharmacokinetic studies of ferulic acid in human plasma. The results show that:(1) Dionex C18column (4.6mm×250mm, id,5μm), methanol-1%glacial acetic acid as the mobile phase, flow rate1.0mL/min, control column temperature was30℃, the injection volume was20μL, DAD diode array detector wavelength is323nm.In the chromatographic conditions, the low limit of detection for the ferulic acid is0.03μg/mL ferulic acid standard solution get a good linear relationship in the concentration range of10~500μg/mL.Linear regression equation is:y148590.7194X-490.00171(R=0.9999).(2) HPLC was successfully used for simultaneous detection of ferulic acid in Radix Angelicae Sinensis, Rhizoma Sparganii and Rhizoma Ligustici Chuanxiong. Target peak are well separated with other impurity peaks. Ferulic acid’content is23.6μg/mL in Radix Angelicae Sinensis,104μg/mL in Rhizoma Sparganii and57μg/mL in Rhizoma Ligustici Chuanxiong.(3) The detection of ferulic acid in human plasma can be completed in13minutes as methanol-0.1%phosphoric acid being the mobile phase.The method then successfully used in pharmacokinetic studies.(4) The limits of POPs are:0.024~0.358ng/mL for OCPs and0.130~0.405ng/mL for PCBs using GC-ECD;20~400ng/mL for OCPs and300~500ng/mL for PCBs using GC-MS. The comparison showed that GC-ECD has higher sensitivity and is more suitable for the detection of trace amounts of chlorinated organic pollutants in the environment.
