Method of using magnetism character of Fe3O4is a important way of manufacturing targeting drug, and the aim targeting in three directions:Nano-Fe3O4’s toxicity towards tumor, Nano-Fe3O4carrying tumor drug itself, by manufacturing water-soluble magnetic carriers to bearing tumor drug. The third has the brightest prospective above these three methods.However,many of water-soluble Nano-Fe3O4manufactured by researchers is covered by high polymers,which broads the diameter of nanometer materials.Therefore, this paper researched dopamine molecule’s modification for drug carrier system.The experiment used Nano-Fe3O4as core to carry quercetin which has the function of suppressing increment of tumer, and modified dopamine to get better water-soluble nanometer material to conquer the drawback that quercetin hardly dissolving in water.Moreover,dopamine has several receptors on the surface of tumor, which increased it’s targeting and bio-solution ability.This report preparared nano-ferriferrous oxide by using FeCl3·6H2O and FeSO4·7H2O as raw material by traditional co-precipitation method.After reaction condition optimization, the better results are followed,dissolving FeCl3·6H2O(aq) and FeSO4·7H2O(aq) with water to keep Fe2+and Fe3+in0.115mol/L,respectively.After heated to80℃and stirred,28%ammonia was added until pH value is9-10.After stirred for another20mins, nano-ferriferrous oxide can be obtained by magnet. It can be confirmed that the black solid is ferriferrous oxide with diameter of25nm by IR,XRD,SEM and TEM.We furtherly gained ferriferrous oxide-quercetin complex by emulsion-coacervation method.After optimization, the best results are followed:mquercetin:mnano-ferriferrous oxide=2:1, methanol, and16hours in room temperature.The size of synthesized nano-ferriferrous oxide-quercetinis is25nm~50nm.Based on the design of free ferriferrous connected with N atom of dopamine, nano ferriferrous oxide-quercetin-dopamine ternary complex had been prepared. The optimized condition is20mL ethanol,10mL tris,20mg nano-ferriferrous oxide-quercetin and15mL dopamine hydrochloride solution, stirred20h in room temperature, separation by magnetic. The size of targeted nano-ferriferrous oxide-quercetinis-dopamine complex is25nm-50nm.The antitumor activity assays in vitro showed that the nano-Fe3O4--quercetin-dopamine complexes indicated moderate anti-proliferation activities against MDA-MB-231and PC3cancer cell lines.
