As a new type of carbon nanomaterials, Fullerenes have a wide range of applications in disciplinary due to the unique chemical and physical properties. But fullerenes only have a very limited solubility in water because of its hydrophobic, and therefore its future applications in biological field are hindered. Recently, it has been found that a good way to improve the solubility of fullerenes in water is introduce hydroxyls groups to fullerenes to produce fullerenol,in this way, fullerenes can also retained fullerenes’unique biological activity, such as antioxidant properties, free radical remover, cell protection and so on.In this thesis, one of the common water-soluble fullerenes’derivatives-fullerenol has been produced by ammonium hydroxide of tetrabutylammonium catalytic synthesis method, and characterized by the method of FTIR, NMR and time-of-flight MS. Then we used fullerenol role in the D-galactiose-inducible mouse spleen cells and co-cultured together, we also observed the survival rate, free radical remover ability of mouse spleen cells’and so on. The following are the main results obtained in this paper:(1) Fullerenol with a good the solubility has been prepared, and it applicable to biological cell experiments.(2) Most of the mouse spleen cells spherical in shape, and then we using trypan blue staining method identified the survival rate and found a high survival rate of the fresh mouse spleen cells. And Establish a new aging spleen cell model by D-galactose with different concentration, through thiazolyl blue (MTT) assay, cell counting method choosing the concentration of10g/l D-galactose to explore Fullerenol’effect.(3) The test results show that fullerenol was added to the cell culture medium to ma ke a final concentration of0.7mg/ml. Viability of spleen cells after induced by D-galactos e was assessed by MTT assay, the result shows that the fullerenol could decreased the cell s mortality rate, survival rate of senescence was69.7%, while the survival rate of spleen cel Is added with fullerenol was80.9%. The activity of MDA and SOD was assayed by MDA d etection kit and SOD detection kit. SOD value of fullerenol is7.7451±0.1064U/mg·pro t, SOD value of D-galactose is4.4362±0.2643U/mg·prot, the result showed that the SO D activity of fullerenol groups kept relatively high activity, which evidence that fullerenol is able to remove free radical in aging spleen cells, so fullerenol has antioxidant prope rties. MDA value of fullerenol is2.34±0.15nmol/mg·prot, MDA value of D-galactose is2.71±0.05nmol/mg·prot, the MDA is lower than the comparison improve that fullere nol reduce the generation of harmful substances in the spleen cells to protect the spl een cells.
