Fluorescent properties of6kinds of camptothecine compounds were studied in this thesis.Relationship between fluorescence and molecular structure was discussed. According to thedifferences between fluorescent properties of camptothecin and10-hydroxycamptothecin, afluorometric method for the determination of10-hydroxycamptothecine in Chinese herbalmedicine common Camptotheca fruit (Xishuguo) was proposed, and the result was verifiedby a thin layer chromatography-fluorescence scanning method.Two parts were included in the thesis:1. Fluorescent properties of9-methoxycamptothecine,10-methoxycamptothecine,9-aminocamptothecin,9-nitrocamptothecin,7-ethylcamptothecin,7-ethyl-10-hydroxycam-ptothecin were studied.9-nitrocamptothecin had no fluorescence and cannot be sensitized toproduce fluorescence.9-aminocamptothecin had fluorescence in aqueous solution with acidityabout pH2.3. Other4kinds have strong fluorescence in the range of weak acidic to weakalkaline pHs. The maximum excitation and emission wavelength λex/λemof9-methoxycamptothecine was350nm/530nm, fluorescence quantum yields in10℅CH3OHsolution and in100℅CH3OH were0.034and0.13, respectively. The λex/λemof10-methoxycamptothecine was376nm/425nm, quantum yield in10℅CH3OH solution was0.95. The λex/λemof9-aminocamptothecin was368nm/453nm, quantum yield in10℅CH3OH solution with pH2.0was0.31. The λex/λemof7-ethylcamptothecin was367nm/420nm, quantum yield in10℅CH3OH solution was0.97. The λex/λemof7-ethyl-10-hydroxycam-ptothecin was376nm/540nm, quantum yield in60℅CH3OH solution with pH13.0was0.20.2. According to the special properties that10-hydroxycamptothecin has strongfluorescence in0.1mol/L NaOH solution but camptothecin has no fluorescence under thiscondition, a fluorometric method for selective determination of10-hydroxycamptothecine inXishuguo was established, and the result was0.0157%. The reliability of the new method wasverified by a TLC fluorescence scanning method, and the result was0.0156%. |
