The deer is precious and can be used for medicine and edible.China is the first country to use deer products to cure diseases.The Compendium of Materia Medica in the Ming Dynasty had a more detailed account of the use of raising deer and deer: "three or five deer were domesticated with three or five newly born deer." In Chinese Pharmacopoeia 2015,the Chinese medicinal materials based on deer include antlers(deer Linnaeus or sika deer Temminck sawing off antlers that fall off in February and May of the following year,or horns that have been ossified).Pilose antler(Temminck,male deer or sika deer of the family Cervus Linnaeus,has not yet ossified young horns).antler cream(where the antlers have been removed from colloidal antlers),Antler glue(solid glue concentrated by antlers),etc.In addition to the above,Luchan is also an important Chinese medicine based on deer,used in Xinnaokang capsule,Xinnaokang tablets and other traditional Chinese medicine prescription.In this study,based on the PCR reaction fluorescence detection technique,a method was established to identify the genuine and false Chinese medicinal materials of deer origin,which can accurately identify the traditional Chinese medicine and its forgeries.Taking Xinnaokang capsule as an example,this paper analyzes and detects the prescription of traditional Chinese medicine containing the ingredients of deer origin,and accurately grasps the actual situation of the ingredients of deer origin in the preparation of traditional Chinese medicine.The annealing temperature and cycle times involved in the PCR reaction system were optimized in different degrees,and the reaction conditions and reaction system were determined.The reaction conditions are as follows: predenaturation at 94 ? for 5 min,amplification reaction at 94 ? for 30 s,reaction for 63 ?,30 cycles for 30 cycles,and extension of 5min,PCR reaction system at 72 ?.As follows: 2 × master mix:12.5 ? L primer F: 1 ? L primer R: 1 ? LddH2O:8.5 ? L 5min.PCR: 2 ? L;The total reaction system: the product of 25 ? L.The concentration of PCR reaction products using agarose gel electrophoresis was 1.5 %.9v/cm gel electrophoresis,until the bromophenol blue indicator migrated to the middle of the gel.Gel imaging system Observe and keep records.As a result of the process,DNA in antler gum was destroyed and could not be extracted and PCR reaction could not be carried out.The sensitivity of PCR reaction fluorimetry was higher than that of other methods,and the lowest detection amount was 0.5%.The issue of food safety has always been the focus of attention of the national economy and people's livelihood.Deer meat is more expensive because of its nourishing effect.Driven by profit,cheap horse meat is flooded in the market,pork and even chicken and duck meat impersonating venison.Regulators urgently need to improve the relevant inspection system to provide strong scientific basis and technical support for market supervision.In this study,a PCR reaction fluorescence detection system for the identification of deer origin food authenticity was established.The detection results of several declared deer meat products on the market show that this method can be used to identify deer food and its counterfeit products.The method is simple,rapid,accurate and the lowest detection quantity is 0.5 %.
