Medicinal ingredients in antler has brought wide attention. This paper comparesthe efficiency of PEF with ultrasonic method for antler water-soluble polysaccharide,and optimize the extraction conditions. Polysaccharide in antler is GAG, also namedacid mucopolysaccharide. Ethanol precipitation and Sevage have been chosen to getGAG, this method own minimum loss and maximum efficiency. We developed abeverage contain polysaccharide of antler. Specific research results are as follows:1. Study on the extraction of water-soluble polysaccharide in antler by ultrasonictechnology and the optimization of the technology.Choose ultrasonic technology as the method of extraction of water-solublepolysaccharide in antler, stationary other factors of ultrasonic processing. by singlefactor test, antler polysaccharide extraction rate as the index, determine the optimalmaterial liquid ratio is12, papain added1.5%and neutral protease added1.5%,ultrasonic power is200W, ultrasonic temperature is40℃, time of ultrasonic is40min.Ultrasonic technology conditions be optimized by response surface method, themathematical model of regression equation is as follows:R=5.18+0.072A+0.15B+1.21C+0.080D-0.12AB+0.15AC+0.48AD+0.045BC-0.45BD+0.017CD-0.45A2-0.58B2-0.64C2-0.54D2Through the analysis and coded values be calculated, the best combination is:ultrasonic power209W, material liquid ratio is12.08, the temperature is49.9℃,ultrasonic time is42.3min. Therefore, Sevage method bechosen to remove protein and supplemented by ethanol precipitation. Peptides solublein70%ethanol, but polysaccharides can not soluble in70%ethanol. The next step isstudy on the separation of the polysaccharides using ion exchange resinDEAE-Sephadex, we gain GAG1, GAG2, GAG3.In this research, antioxidant activity of GAGs was detected, superoxide anion、DPPH free radicals clearance and hydroxyl free radical clearance will be compared.When the concentration of GAG1is0.25mg/mL, the superoxide anion free radicalclearance rate reached6%, the DPPH free radical clearance rate reached12%, thehydroxyl free radical clearance rate reached57%. When the concentration of GAG2is0.25mg/mL, the superoxide anion free radical clearance rate reached12%, the DPPHfree radical clearance rate reached28%, the hydroxyl free radical clearance ratereached61%. When the concentration of GAG3is0.25mg/mL, the superoxide anionfree radical clearance rate reached4%, the DPPH free radical clearance rate reached8%, the hydroxyl free radical clearance rate reached33%.4. Study on hawthorn and antler polysaccharide antioxidant nutrition beverageDevelop the hawthorn and antler polysaccharide antioxidant drinks use higheractivity GAG extracted before. SSPS and CMC-Na were chosen as stabilizer. By thesingle factor and orthogonal test, the test results showed that0.12%of SSPS mixedwith0.25%of CMC-Na is better. On the choice of sweeteners, the results showed thatwhen compound2%sucrose and4%xylitol as sensory, the score is the highest. Thesterilization conditions: sterilization temperature121℃, sterilization time is10min.Drinks can store more than half a year.Through the study on the beverage traits, health indicators, determination ofphysical and chemical indicators, microbial indicators, the beverage industrystandards can be determined. Finally through fuzzy evaluation to classification of thebeverage, we gained93.87points, achieved a good standard.
