Study On Extraction, Separation, Purification And Antioxidant Activity Of Total Flavones From Apricot Blossom | | Posted on:2011-05-12 | Degree:Master | Type:Thesis | | Country:China | Candidate:J J Yu | Full Text:PDF | | GTID:2131330332470454 | Subject:Food Science | | Abstract/Summary: | PDF Full Text Request | | This paper study the technology condition on extraction methods of total flavones from apricot blossom about microwave-assisted and water-bath.Dry apricot blossom was used as raw material,alcohol concentration as lixiviation solvent and extraction content as evaluation index.Separation and purification of total flavones from apricot blossom were initial performed on silica gel column chromatography.Through the DPPH free radial elimination system and the O2- free radial elimination system,the antioxidant activity and the free radial elimination ability of total flavones from apricot blossom were evaluated.The conclusion as follows:(1)The optimum technology parameters of microwave-assisted extraction of total flavones from apricot blossom:alcohol concentration 60%,irradiated time 8min,material-liquid rate 1:80,irradiated temperature 70℃,on this condition of extraction total flavones content is 210.48 mg/g.The influence of four factors on the extracted was that alcohol concentration> irradiated time> irradiated temperature> material-liquid rate.(2)The excellent and economic technology parameters of water-bath extraction of total flavones from apricot blossom:alcohol concentration 50%, terial-liquid rate 1:30, extraction temperature 70℃,extraction time 2h,on this condition of extraction total flavones content is 175.70mg/g.The influence of four factors on the extracted was that extraction time> alcohol concentration> material-liquid rate> extraction temperature.(3)compared the method of microwave-assisted extraction with the method of water-bath extraction, the method of microwave-assisted extraction have more efficiency,time-saving,energy-saving,simple technology,less dopant,more prospects of exploitation and application.(4)The separation and purification of total flavones from apricot blossom were identified by color reaction,thin-layer chromatography(TLC),silica gel column chromatography,ultraviolet spectra(UV),ultra performance liquid chromatography(UPLC),time-of-flight mass spectrometry(TOF MS).Study results indicate that:the total flavones from apricot blossom in acetic ether abstraction have abound with flavone,flavonol and isoflavone.(5)Through the DPPH free radial elimination system and the O2- free radial elimination system,antioxidant activity and the free radial elimination ability of total flavones from apricot blossom were evaluated.The results indicate that:the total flavones from apricot blossom as a natural antioxidant of exploitation value that have invariably eliminated ability of DPPH free radial and O2- free radial. | | Keywords/Search Tags: | apricot blossom, total flavones, extracted technology, separation, purification, antioxidant | PDF Full Text Request | Related items |
