With the improvement of people ’s living conditions,cosmetic has become thenecessary of everyday life,the quality and safety of cosmetic products is closelyrelated to people’s lives. Our country has placed great importance on cosmetic safety,and therefore drew up many related laws and regulations, but there are still someimperfections of method for additive agent from cosmetics and their packaging. Themain ingredients of cosmetics are oil and wax, which will easily deteriorate during theprocess of production and use, there are some antioxidant added usually in order toensure the qulity of cosmetics,prevent oxidation, butyl hydroxyanisol, BHA and butylhydroxyl roluence, BHT are both common antioxidant. Excess BHA and BHT areharmful to health, the national maximal limit for BHA and BHT according to thenational standard “Hygienic Standard for Cosmetic”(2007) is0.15%.The packaging of cosmetic has the functions of hold and protect during thetransport, for the convenience of cosmetic storage, playing a important role in thecosmetic sales, is a inseparable component of cosmetic. Glass and plastic are bothcurrently in common use, the ingredients of cosmetic plastic packaging are polythene,polypropylene, polyethylene terephthalate and so on. The packaging will be transparent,durable, light, anti-impact and prevent acid material from corroding the out packaging,when the BPA is added during the production processing of plastic. BPA has a similarchemical structure to estrogen, possessing weak estrogen activity and strongantiandrogens activity,being able to interfere with regular endocrine function, eitherdirectly or through other biological derivant. Bisphenol A has been added into theblack list of prior pollutants in some countries, the European Union has banned theproduction of baby bottles which contains bisphenol A since March2,2011, People’sRepublic of China Ministry of Health, No.15, Notice of year2011has also made aseries of requirements the use of BPA. But it is currently rare reported about thestandard, specification and testing method for Bisphenol A from the cosmeticpackaging. To make precise, rapid and proper testing method for the analysis and testing ofantioxidant from cosmetic as well as the BPA from cosmetic plastic packaging is ofgreat significance for the protection of human beings’health.Objective:To establish the method for detecting BHA and BHT in the cosmetic and BPA inthe cosmetic plastic packaging with liquid chromatography tandem mass spectrometry.It has completed the detection of BHA and BHT of30types of cosmetic as well as theBPA of30types of packaging for cosmetic.Methods:In this paper, using Ultrasonic to extracted the BHA and BHT in the cosmeticsamples and investigating the influences of time and extraction solvent on theextraction rate for BHA and BHT in the cosmetic.The BPA would be extracted from the cosmetic samples by using the UltrasonicExtractions Method, investigating the influences of extraction solvent and time on theextraction rate for BPA in the cosmetic packaging.The extracts from cosmetic and the packaging samples would be purified byusing the Solid Phase Extraction method, investigating various Solid Phase Extractioncolumns, eluent and the influence of eluent volume on the purifying effect of theextracts from the cosmetic and the cosmetic packaging samples.The BHA, BHT and BPA from the samples would be analyzed by using the liquidchromatography tandem mass spectrometry method, to raise the accuracies for therecovery test along with the verification methods of precision degree.Results:When Methanol was adopted as the extraction solvent, extraction time was30minutes, extraction frequencies were twice, the extraction rate for BHA and BHTfrom3categories of cosmetics all exceeded91%.When Hexane was adopted as the extraction solvent, extraction time was20minutes, extraction frequencies were twice, the extraction rate for BPA exceeded92.4%.When PLS was adopted as solid phase extraction column,using methanol aseluent, the volume of eluent was6ml,the purifying effect of BHA and BHT fromcosmetics and BPA from the cosmetic packaging were better,the recovery rates were95.3%,94.1%and93.2%. When liquid chromatography tandem mass spectrometry was used as method oftesting for BHA and BHT from cosmetic, showed that the recovery rate of BHA was86.0%~98.0%, Relative Standard Deviation(RSD)was2.3%~6.1%, the recoveryrate of BHT is84.9%~97.4%, Relative Standard Deviation(RSD)was2.1%~6.5%.BHA and BHT had better linear relationship within the range1ng/mL to500ng/mL;correlation coefficient was greater than0.9996.When liquid chromatography tandem mass spectrometry was used as method oftesting for BPA from cosmetic plastic packaging, the recovery rate of blank samplesand the BPA recovery rate is85.1%~94.5%, RSD is1.7%~6.48%. BPA has a betterlinear relationship within the concentrations range of1to500ng/ml, the correlationcoefficient is0.9999.Conclusion:This research has established the method for detecting BHA and BHT in thecosmetic and BPA in the cosmetic plastic packaging with the liquid chromatographytandem mass spectrometry. It is confirming the extraction condition and purificationcondition of solid phase extraction for BHA, BHT and BPA from the samples. Thedetection limit of the method for BHA and BHT from the established cosmetic is2ng/g in this research, lower than the national standard (0.05mg/g) according to theGTB22728-2008“Determination of Butylated hydroxyanisole (BHA) and Butylatedhydroxytoluene (BHT) in cosmetics-High performance liquid chromatography”, asthe innovation of this research.The detection limit of BPA is1ng/g in cosmetic plastic packaging sample,Thismethod has reference value for the the establishment of testing standard for BPA fromcosmetic plastic packaging as a result of this lower detection limit.By adopting the established methods in this research, the result of testing forBHA and BHT from30various cosmetic of3categories-cream, emulsion and lotionand the BPA from30kinds of cosmetic plastic packaging shows that: there were13kinds of cosmetic contains BHA,18kinds of them contains BHT, but they all did notexceed the national standard;Only5types,which contained BPA, were detectedfrom all the30kinds of cosmetic plastic packaging.
