Momordica Charantia L was a tropical and subtropical plant for edible and medicinal. Momordica Charantia L contains triterpenoids, polysaccharides, protein, amino acids, fatty acids and other active substances. Pharmacological studies have shown various pharmacological and biological activities including hypoglycemic, lipid-lowering, anti-oxidation, anti-cancer, hepatoprotective and other biological activity. In Chinese folk, Momordica Charantia L is widely used as jianghuo and clearing heat plant, while the mechanism of jianghuo has not yet been reported. And it has been made into different products. It also needs further intensive processing. In order to compare the quality and function evaluation of different drying methods on Momordica Charantia L, this paper examines six different kinds of dried Momordica Charantia L; the restraint stress as a model to study the mechanisms of Momordica Charantia L improvement effect on liver injury and lipid metabolism disorders. The main results are as follows:1Effect of different drying methods on physicochemical property of Momordica Charantia L: Effects of drying methods of FVD, MD, HPD, VD, SD and HAD on physicochemical quality of Momordica Charantia L was studied. Comparison results showed that there was similar changing trend of polyphenols and flavonoids content, which was HAD>MD>HPD>VD>SD>FVD. The ORAC antioxidant capacity was HPD>VD≈MD≈HAD>SD>FVD. There was significant difference between ORAC and the content of polyphenols and flavonoids. For different drying methods, there was a correlation between sensory evaluation and sensory color. There was also a big difference of surface and cross-sectional structures of dried Momordica Charantia L. Effect of different drying methods on volatile components of Momordica Charantia L:For Momordica Charantia L dried slice, there were194volatiles, of which16were in all the samples. While the species of Momordica Charantia L tea was119, of which3were in all (2-hexenal,(E,E)-2,4-heptadienal, phenylacetaldehyde.). The result of principal components analysis showed that (E,E)-2,4-heptadienal, benzaldehyde, nonanal, hexanal,3-methyl-pentadecane, hexadecane, cis-2-pentenyl furan,2-n-pentyl furan were the main volatile components in Momordica Charantia L dried slice2The protective effect and mechanism of Momordica Charantia L on restraint-stress-induced liver injury:The6weeks old male mice were randomly divided into5groups including normal control group, stress control group, three MCFE groups (250mg/kg,500mg/kg and750mg/kg), which were administered samples once a day successively for7days. Using HE staining method to observe the morphological changes of liver. The alanine aminotransferase (ALT), aspartate aminotransferase (AST) activities, malondialdehyde (MDA) of serum were measured by colorimetric method. The glutathione (GSH), malondialdehyde (MDA), NO content and the activity of glutathione peoxidase (GSH-Px), total superoxide dismutase (T-SOD), inducible nitric oxide synthase (iNOS) in liver were also measured by colorimetric method. The expression of iNOS protein was analyzed by using Western blot method. The antioxidant capacity was measured by oxygen radical absorbance capacity(ORAC)assay. The results shows that: Compared with the model group, MCW significant reduced the ALT and AST activity in serum (p<0.05). Medium and high treatmeat groups could increased GSH content in liver; the SOD activity in liver of high treatmeat group were significantly increased (p<0.05); The GSH-Px increased insignificantly; MDA content in serum and liver of each treatmeat groups were significantly reduced (p<0.05); ORAC in serum and liver of each treatmeat groups were significantly increased(p<0.05); The NO content and the iNOS activity of liver were significantly reduced (p<0.05); the ROS content in mitochondria of liver was significantly reduced (p<0.01), mitochondria membrane potential of liver was significantly increased (p<0.05); the respiratory chain complex I and II in liver of each treatmeat groups ware significantly increased (p<0.05). Each treatmeat groups also could improve hepatic morphologic.3The protective effect of Momordica Charantia L on serum lipids metabolic disorder in restraint mice:The6weeks old female mice were randomly divided into5groups including normal control group, stress control group, three MCW groups (250mg/kg,500mg/kg and750mg/kg), which were administered samples once a day successively for7days. After intravenously injected10%Intralipids into the20h restrained mice, the content of serum lipid, lipase of mesenteric adipose tissue (MAT) and lipoprotein lipase (LPL) activity of different tissues were determined. The oxidative situation and the antioxidant capacity of the body were measured. Compared with the control groups, MCW accelerated the elimination rate of serum triglyceride (TG), improved the lipase and LPL activity of mesenteric adipose. And it also decreased the level of MDA and increased the antioxidant capacity of serum (ORAC)...
