Bitter gourd(Latin name:Momordica L.)belongs to the Curcubitaceae family.It has other names:liang guajinlizhi,laiputao etc.It has many chemical compositions.The saponions is one of the mainly active ingredients from bitter gourd. It is reported that the saponins from bitter gourd have been used for the treatment of hyperglycemia, high blood-fat, tumor AIDS, anti-phloggistic, immune function to human being etc.In this thesis, our research focuses on extraction, fractionation and isolation of saponins from Momordica Charantia L and we also make a study of function of antioxidation in vitro and hypoglycemic activity.Using two methods to extract saponins. one of the method is ultrasonic, the optimum cond-itions of extracting saponins are determined through single factor, PBand orthogonal test. With a ratio of 1:40 between Momordica charantial and 70%ethanol,30min,ultrasonic power is 360W,80℃. Another method is enzymolysis, when the saponins were extracted by enzymolysis method, the optimum conditions of extrating saponins are determined through single factor, PB and CCD test. With 70%ethanol,1:21.55 liquid ratio, an addition of 0.604%. pH5.54, the main active ingredients were extracted at 51.7℃.Four kinds of macroporous resins were selected to compare their absorption and desorpt-ion of saponins. Result showed that AB-8 resins was the optional to purify the saponins from Momordica charantia L. Then the conditions of purification of saponins with macroreticular resin are studied by static and dynamic experiment. The parameters of dynamic adsorption are as following:the original concentration 5mg/mL, the feeding rate 2BV/h and pH value 10; The parameters of dynamic elution are as following:the eluting cubage of 80%alcohol, the eluting rate is 2BV/h, the eluting volumn is 4.5BV.The antioxidation in vitro and hypoglycemic activity effect of saponins from Momordica Charantia were studied. The resulted showed that:the scavenging capabilities to superoxide anion radial, the hydroxyl free radical and DPPH radical were well, and also the saponion of Momordica charantia with alcohol could have the activity of agitating for IRTK. Then it provides the basis for designing the lead compound of new drugs for treating diabetes.
