In recent years, with the deepening of the research, the varieties of utility of fenugreek plant caused extensive concern in different industries both at home and abroad. It has been hailed as " Every Part of it Precious " and it is one of the eight major economic crops in the world.The composition of the Fenugreek seeds contain galactomannan, trigonelline, diosgenin,4-hydroxyl isoleucine, which all have high economic value and huge social demand.The galactomannan in Fenugreek distributed in the endosperm of seeds, people failed to make substantive breakthrough in some aspects such as the key technology of peeling off the skin and the separation of cotyledon and endosperm for many years.The application of the research projects in domestic focused primarily on the separation and purification of two or three components in fenugreek. So far, no research achievement had been found to develop a reasonable technology route to make it achieve the industrialized production, the key technologies breakthrough of Fenugreek means that the era of comprehensive utilization of High Additional-value of Fenugreek is coming, which possess immeasurable value in economic, social and environmentalFor the above questions, there is a urgent need for us to solve the practical problems in the society, This topic carry ed out study of preparation technology about fenugreek gum, fenugreek oil and diosgenin and other groups of small molecules, In order to achieve the comprehensive and effective utilization of fenugreek resources, and improve the social and economic benefits. The result was as follows:1.A research and optimization to prepare the fenugreek gum had been established. On the basis of single factor experiment to choose concentration of sodium chloride solution, material liquid ratio, soaking temperature and soaking time as inspection factors, regarding peeling ratio as index while quantification for the index, and designing four factors three levels orthogonal experiment We came to the conclusion that the optimal peeling process conditions for sodium chloride solution concentration was3%,material liquid ratio was1:15,the soaking time was25min, soaking temperature was100℃by analysing experimental data.And different preparation methods had been developed on the effects of the molecular weight of fenugreek gum and dissolution rate.Results showed that the sticky molecular weight of primary gum was M=4.9x10s, and for alcohol precipitation gum M=6.5×105, the sticky molecular weight of fenugreek gum increased obviously after the water soluble alcohol sank.The dissolution time of alcohol precipitated gum powder is2.6times of the original powder,but the viscosity of colloidal sol had been rised by675.84%.2.A research to prepare fenugreek oil and diosgenin in fenugreek had been established. By adopting the combination of single factor with orthogonal experiment method, regarding diosgenin yield as index while quantification of the index. The experimental results are as follows: extraction time was6h,60-80mesh size, oil extraction ratio was8.98%. Acidolysis condition was as follows:sulphuric acid concentration was4%, reflux time was5h, acidolysis temperature is70℃, suitable particle size is24to40meshes, the extraction yield of diosgenin under the above condition was0.66%.3.A research about the extraction process of trigonelline had been launched. On the basis of single factor experiment to choose solvent type, solid-liquid ratio, ultrasonic power, time as the influence factors and optimize orthogonal designing. The experimental results are as follows: extractcted by water, material liquid ration was1:60, ultrasonic power was90w, ultrasonic time was30min. The results show that the technology was feasible.4.A method for the determination of trigonelline in Fenugreek by HPLC had been developed. The chromatographic experimental conditions were as follows, Venusil ASB C18(4.6mmx250mm,5μm); Mobile phase:methanol-water (1:1), Flow rate:1.0ml/min; Column temperature:room temperature;Sample quantity:10μl; the detection of wavelength by UV:265nm; the standard curve was linear in the range of0.10-2.00ug,r=0.9993(n=5).The average recovery rate was100.00%, RSD=1.78%. The result of determination of trigonelline in Fenugreek from Tongxin, Ningxia was3.3mg/g。5.Taking defatted fenugreek cotyledons as raw material, extraction and detection methods were studied on its effective componenis—4-hydroxyl isoleucine. On the basis of single factor experiment to choose ethanol concentration, material-liquid ratio, ultrasonic power, time as the influence factors and optimize orthogonal design. The experimental results are as follows:20%ethanol, material-liquid ratio was1:70, ultrasonic power was120w, ultrasonic time was40min,4-hydroxyl isoleucine extraction yield of0.51%under the above conditions. Determine the content of formaldehyde by the formaldehyde derivative method. The results revealed that the technology was feasible. It has a certain reference value for prompting the extraction of4-hydroxyl isoleucine industrialize.
