Use modem science and technology and research methods extract the active chemicalcomposition from the herbal, People can make efficient use and little or no toxic side effectsof natural drugs, Chinese herbal medicine to be able to play to the advantages. Fenugreek is acommonly used Chinese herbs, a rich activity of the original chemicalcomposition—Protodiosein. Protodiosein belonged steroidal saponins (steroidal saponins),also known as furans steroid saponins, has strong biological activity, such as enhancementof male sexual function, kill many cancer cells: hypolipidemic and anti-leukocyte function.To establish a method of HPLC determination of the content of protodioscin estractedfrom fenugreek, on the basis of the way the predecessor estracet it fromherbal, chromatographic conditions: Lichrospher C18 column (4.6mm×250mm, 5μm), themobile phase: acetonitrile-water (8: 92), the column temperature: 30℃, flow rate:0.5mL/min, the detection wavelength: 203nm.To use established HPLC method for the ultrasound extraction research of protodioscinfrom ethanol solution, and determine the best ultrasonic extraction technology throughorthogonal design, the best extraction process: Extraction of 75% ethanol solution, the feedratio of 1: 60 (M/V), ultrasonic extraction three times, each time for 15 minutes.Macroporous resin with good adsorption is a class of organic polymer adsorbentdeveloped in the 60 years, 20th century. To study the best purification condition fromfenugreek by adopting macroporous adsorption resin. The optimum purification conditionswere as follows: HP-20 macroporous adsorption resin was selected as the adsorbed resin, theratio of resin to crude drug was 4: 1, the ratio of diameter to height of resin column was 1:12. the first step is use water to wash away the impurities by the flow speed of 3BV/h, and70% alcohol was chosen as elution agent, the flow speed of adsorption was 4BV/h, the flowspeed of cleaning and elution agent was 10 BV/h.
