Glutathione peroxidase (GPX) Selenium is an important antioxidant enzymemimics, hydrogen peroxide can be used to restore GSH. ROS body maintainhomeostasis, so that the body against oxidative damage. A variety of diseases areassociated with ROS imbalance can not be timely removal of ROS attackbiomolecules lipids, proteins and DNA. leading to mutations and apoptosis. knownsomatic mutations in the gene cause the root of all cancer, extensive research hasproved that the redox imbalance is one of the main reasons induced carcinogenesis.So for one analog GPX antioxidant enzymes, it is particularly impotrant, not only tohelp the study of the structure and catalytic mechanism of natural GPX. but also forthe future development and clinical application of these drugs provide anexpeirmental basis.Although the active peptide sequence is short less the number of amino acids,but with all the conditions of play activity, in addition, small peptides in vitro to forma stable solution can not be conformation-speciifc function and to the substratesufficiently. Through structural analysis and compairson of GPX superfamilv foundaround the catalytic center located Sec Gln79and Trpl53in the conservative position.and therefore, presumably has an extremely important role to play when the catalyticfunction of these three amino acids in the natural GPX. This paper attempts to useFmoc solid-phase peptide synthesis method Sec. Gin and Trp three amino acidtirpeptide UWQ, which Sec as active sites. Gin and Trp as a secondary amino acids,the determination. GPX activity of the tirpeptide is2.10U umol. Late stage lungcancer cell model established to explore the impact of UWQ on lung cancer cellgrowth and migration, the study results show that UWQ HepG2cells can effectivelyinhibit the growth and proliferation of HepG2treated by UWQ significantly-decreased ability to heal scratches, cell motility dysfunction. In addition. UWQ cansignificantly reduce MMP-9and AFP, thereby reducing the ability of invasion andmetastasis of liver cancer cells. This paper studies obtained tripeptide LJWQ and its impact on lung cancer cellsas GPX enzyme development and simulation of these drugs provide an experimentalbasis and clinical applications in vivo animal.
