Ibuprofen, a non-steroidal anti-inflammatory drug, can alleviate acute or chronic rheumatoid arthritis, osteoarthritis, ankylosing spondylitis and soft tissue rheumatism attack. It is used to treat mild or moderate pain such as dental, obstetrics, orthopaedic surgery pain and pain caused by sports injury of soft tissue of the muscles, bones. It also can relieve headaches and pains of non-rheumatoid arthritis of inflammatory bursitis and tendinitis. It has analgesic and anti-inflammatory effects for gout. It can also be used for high fever caused by the common cold or influenza. Ibuprofen has been more favored by consumers since the advent, because it has stronger effects onanti-inflammatory, pain-relief and defervescence than aspirin, phenylbutazone and acetaminophen.The synthetic processes of Ibuprofen mainly include the traditional Boots process and the latest BHC green synthesis process. After many years of exploration, Shandong Xinhua has adopted the improved synthesis process:using the isobutyl benzene as starting material and adopting1,2-dislocation rearrangement process. The synthesis process of Ibuprofen included6steps, i.e. Chloroketone, Condensation, Rearrangement, Hydrolysis, Acidification and Refining.Class1solvent dichloroethane was used in the step of chloroketone during the manufacturing process of Ibuprofen, but dichloroethane was prohibited from using by EDQM. About70%Ibuprofen was used for export, which implied that our Ibuprofen couldn’t export to Europe if we didn’t change the solvent dichloroethane. The solvent Methylene chloride replacing dichloroethane was used in the step of chloroketone by many trials. The yields of each step and total yields of Ibuprofen were basically identical to those of the original process, and the quality were better than the original process, met the requirements of USP, BP and EP6.1, which provided a quality guarantee for Ibuprofen smoothly distributed to European and American market.The condensation required a long time to react completely, which decided the production period of Ibuprofen, so shortening the condensation reaction time could improve the production efficiency, reduce the cost of raw materials and energy consumption. In addition, we investigated the effect of reaction temperature, reaction time and quantities of catalyst used in the arrangement step on the manufacturing process of Ibuprofen by orthogonal experiment, and optimized the original manufacturing process, found the optimal process conditions, improved the productivity and reduced the running cost.The research results and industrial application showed that the optimal manufacturing process not only improved the quality of Ibuprofen, stabilized the product yields but also shortened the production period of Ibuprofen, reduced the cost of materials and saved the energy. The effects of the manufacturing process were remarkable, the running cost was low and produced some economic benefits, had high value of popularization.
