Coriolus versicolor is a kind of fungi parasitizing on the withered trees and called orgamism protecter. It is said that Coriolus versicolor polysaccharide (CVPS) has strong bioactivity and is giving more and more attention. The paper studied mainly on the ultrasonic assisted extraction of polysaccharide from Coriolus versicolor. and in constrast with the extraction in hot water. At the same time antioxidative activity of CVPS was studied and three constituents of CVPS were obtained by step-Depositing . The main conclusions are as follows:To extract CVPS from Coriolus versicolor with hot water ,the auther revealed the influences of three factors—extracting time, extracting temperature and water-material ratio on extraction rate and purity and optimized extraction technology by RAS and got a better extraction condition: water-material ratio was 55 millilitre per gram, extracting temperature was 80°C,extracting time was 173 minutes. The final extraction rate and purity were 74.38% and 70.92% respectively.To reveal the influences of three factors—alcohol concentration, depositing time, depositing temperature on the yield of CVPS, the auther got a better deposition condition by orthogonal test: elcohol concentration was 80%, depositing time was 4 hours, depositing temperature was 5°C.The yield of CVPS was 4.8%.To extract CVPS from Coriolus versicolor by Ultrasonic assisted extraction and the effect of initial temperature, ultrasonic time, ultrasonic intermission time, mixing speed, ultrasonic power, ultrasonic extracting time and water-material ratio on extraction rate and purity decomposing was studied. On the base of it ,the auther had got a better extraction condition by RAS: the initial temperature is 60°C, pulsed ultrasonic time is 4s, pulsed ultrasonic intermission time is Is, mixing speed is 13r/s, output power of ultrasonic is 1100W,extraction time is 77.5min ,and water-material ratio is 70 millilitre per gram. The final extraction rate is 84.8%.and purity is 83.6%.The experiment studied antioxidative activities of two kinds of polysaccharides gained by the better extraction technology, which including the experiment of clearing DPPH radical, hydroxid radical and superoxide anion radical ,reducing power and Fe2+ chelation activities. The auther had gained the conclusion that CVPS has antioxidative activities and the antioxidative activities of two kinds of polysaccharides are different. Step-Depositing polysaccharides concentrated solution gained by the better Ultrasonic assisted extraction condition with 20%、55%、80% ethanol and the ratio of outcome is CVPS-A: CVPS-B: CVPS-C=32.5%:33.9%:33.6%. CVPS-A’s molecular weight is larger than CVPS-B and CVPS-C. The antioxidative activities of them were studied. The experiment results showed that CVPS-B possesses the best antioxidative effect.
