To make clear the rooting mechanism of cuttings rooting of Alnus rubra, cuttingpropagation technique system and anatomic development of adventitious roots was investigated,proteins expressed during adventitious roots development were extracted and identified usingproteomics technique. The main results were as follows:1The best rooting effect of softwood cuttings of Alnus rubra was reached by treating withGGR-6250mg/L then keeping2h.2The changes of morphology and anatomy were observed during adventitious rootdevelopment, root primordia formed from the vascular cambium parenchyma cell and callus cells,the formation of adventitious root was induce-to-root type.3Some steps of2-DE which is important in proteomics was improved. We also establisheda technique method of2-DE which applied in Alnus rubra:2-DE could be optimized by using themodified TCA/acetone extraction. Totally, When1000μg protein was separated using pH4~7IPG strip, and second dimensional electrophoresis was vertical electrophoresis of12%SDS-PAGE, many protein spots could be well resolved and detected in the gels. All the resultssuggested that the method was suitable for the study.4The function of differentially expressed proteins during key time-point of adventitiousroots development in Alnus rubra were analyzed using MALDI-TOF-MS, result showed thatL1and L3which were close to chitinase can protcet plant from different attack. L4might bequinone oxidoreductase,participated in carbon and energy metabolism. L7was similar to actinwhich can transfer signal.In summary, the formation and development of adventitious roots of Alnus rubra cuttings isa complex process, these results was a basis for study the mechanism of adventitious rootdevelopment, while provided a reference for the other plants.
