Objective to investigate the change of expression and distribution of sIgA, IFN-γ,IL-6protein and sIgA, IFN-γ,IL-6,IL-2,CD3mRNA of macaque’s lung and trachea which were infected with Idiopathic Streptococcus pneumoniae,in order to find more efficiency way to treatment and prophylaxis this morbid.The experiments choice infected Idiovariation Streptococcus pneumoniae macaque for experimental animal, study ing pathological change of lungs,trachea,spleen and lymph node by HE staining,detecting expression of sIgA,IFN-γ,IL-6protein by immunohistochemistry and sIgA, IFN-γ,IL-6,IL-2,CD3mRNA by tissue in sity hybridization in these tissue.results(1) Pathological change of lungs,trachea,spleen and lymph nodeThe lungs in red hepatizaiong period,little serous exudate exudate into pulmonary alveolus.trachea was bleeding,many red cells in ectotrachea connective-tissue and Tracheal cavity. A great number of red cells can be found in red pulp.congestive obviously.The number of lymph node has decreased,paracortex zone turn narrow,some macrophage in medulla.(2) Expressions of sIgA, IFN-γ,IL-6proteinThe expression of IFN-y protein in alveolar septum and pulmonary intravascular in infected group have increased,and significant higher than healthy group(P<0.01),in vessel wall the expression of positive-staining was higher than healthy group(P<0.05);The expression of IFN-γ protein in tracheal muscle and tracheal epithelium in infected group was higher than healthy group(P<0.05);Distribution area of IFN-γ protein in red pulp is lesser(P<0.05);The expression of IFN-γ protein in lymph node in infected group was higher than healthy group(P<0.05).The expression area of IL-6protein in infected group was was lower than healthy group(P<0.05); The optical density of IL-6protein in lymph node was significant higher than healthy group(P<0.01).The expression of sIgA protein positive-staining in lungs of infected group was significantly more than healthy(P<0.01),while lungs intravascular of infected group was higher than healthy group(P<0.05); The expression of sIgA protein in spleen of infected group was significantlty higher than healthy group(P<0.01).(3) Expression of sIgA. IFN-γ、IL-6、IL-2、CD3mRNAThe expression of IFN-ymRNA positive-staining in alveolar septum and lungs intravascular of infected group were significantlty higher than healthy group(P<0.01),in lungs vessel wall was higher than healthy group(P<0.05);The expression of IFN-ymRNA positive-staining in tracheal muscle and tracheal epithelium of infected group was more than healthy group(P<0.05).The expression of IL-2mRNA positive-staining distributed in alveolar septum of infected group and higher than healthy group(P<0.05).Optical density values of IL-6mRNA positive material and expression area in infected all higher than healthy group(P<0.05),positive area of blood in infected group was higher than healthy group(P<0.05);The expression of IL-6mRNA in lamina propria of infected group was significantly higher than healthy group(P<0.01),in tracheal epithelium, expression of IL-6mRNA was higher than healthy group(P<0.05);Distribution of IL-6mRNA positive material in lymph node of infected group was higher than healthy group(P<0.05).Optical density values of sIgA mRNA in lungs vessel wall and blood was significantly higher than healthy group(P<0.01).Distribution area of sIgA mRNA in alveolar septum was higher than healthy group(P<0.05);Tthe optical density values in tracheal epithelium and tracheal muscle of infected group was significantly higher than healthy group(P<0.01),in lamina propria was higher than healthy group(P<0.05).Distribution area in tracheal epithelium of infected group was higher than healthy group(P<0.05);sIgA mRNA immune positive material in spleen of inrected group was higner than healthy group(P<0.05).CD3mRNA immune positive material in spleen of infected group was significantly higher than healthy group(P<0.01);The expression of CD3mRNA in lymph node was higher than healthy group(P<0.05).ConclusionPathological section show that after Streptococcus pneumoniae infection,damage of lungs,trachea was obviously than spleen and lymph node, Main pathological changes is bleeding, congestion and serous infiltrates.each cell factors and antibody in different parts of the organization have different expression strength, IFN-γ、IL-6and slgA were obvious enhancement,IL-2and CD3in infected group have no significant different with healthy group.Every cell factors have connected with each other to made a cytokine network, Mucosa immunization and natural immune cooperated to against the violation of streptococcus pneumoniae.
