Analysis the polymorphism of11micro-satellite DNA seat (AHT4、ASB2、HMS3、HMS7、HTG4、LEX3、LEX074、UM002、UM011、UMNE191、VHL20)that selected from Three horses populations(Kirgiz horses, Russia Trotter, thoroughbred horses), comparing the number of alleles, the average numberof alleles, peptides Information in three populations. blood samples were collected10ml with Sodiumcitrate tube, respectively. Detect the DNA seat of selected11microsatellites with PCR-SSCP in laboratory,and the data analysis with Popgene32computer program. Compare the difference of Ne and allelefrequency distribution among three breeds, and supplement with unique genotype bar for each breeds toconduct analysis of genetic diversity of the three horse breeds.The Ne of microsatellite markers in three horse breeds were significantly different (P <0.05) or highlysignificant difference (P <0.01). it is an important basis for preliminary identification of three horse breeds;On11microsatellite markers for PAGE testing, found that genotypes of ASB2microsatellite-marker inKirgiz horse unique genotype: CC, CD, EG, EH, Thoroughbred unique genotype: BI, JJ, KK; genotypesof VHL20microsatellite-marker in Russian trotter unique genotype: CC,CI; genotypes of AHT4microsatellite-marker in Kirgiz horse unique genotype: DF,EF,GG, Thoroughbred unique genotype: BC;genotypes of HMS3microsatellite-marker in Thoroughbred unique genotype: CC; genotypes of HMS7microsatellite-marker in Thoroughbred unique genotype: AA; genotypes of HTG4microsatellite-marker inRussian trotter unique genotype: BC、BD; The genotypes of UM002microsatellite-marker in Thoroughbredunique genotype: AA; genotypes of UM011microsatellite-marker in Russian trotter unique genotype: AG;genotypes of LEX074microsatellite-marker in Thoroughbred unique genotype: CG. Among the breeds,same microsatellite marker genotypes and allele frequencies there was a significant difference (P <0.01),become another important basis of analysis of genetic diversity.The identification of3horse breeds can be screened by the effective number of alleles (Ne), thedistribution of microsatellite marker allele frequency. Supplement with unique genotype bar for each breedsto conduct analysis of genetic diversity of the three horse breeds.
