Barley mature embryo and anthers Regeneration System in Tibet is still in the blank, Under theenvironment of high altitude, low pressure, dry conditions, this paper selected Dark Blue320, DarkBlue311, Dark Blue148, Himalayan No.4, Himalayas No.6, Dark Blue690, Dark Blue85, LhasaGoumangbai barley, Dark Blue3179, Plateau Zao No.1, Holly8, Holly11, Holly17, Guoluo asalternative materials, using their mature embryos and pollen as explants. Through comparativeexperiments with MS medium and N6medium under different concentrations of2,4-D, ABA, KT,6-BA,IBA, successfully filtered Dark Blue320, Himalayan No.6, Dark Blue85, Lhasa Goumangbai barley,Holly11, Guoluo as appropriate regeneration alternative materials, identified MS medium as matureembryo medium, N6medium as anthers medium, established regeneration system of barley matureembryos and anthers under different concentrations hormones.Tibetan barley breeding has been using conventional breeding methods, high quality varieties selectedbreeding, however, and there exists serious defects which only molecular breeding technique canachieve barley traits improvement in a short time period purposefully. As basic work for barleymolecular breeding, Barley mature embryos and anthers Regeneration System is still under a blank.This paper solved Tibet barley mature embryo and anthers regeneration, offered further preparation oftransgenic barley work and finished experiments of Agrobacterium tumefaciens transformation DarkBlue320and Guoluo.
