Aims:This study is designed to test the digital gene expression profile (DGE)constructed in our lab which was used for comparing the gene differential expression in bone marrow between PRRS model and healthy pigs. Two target genes, NEU1and LAMP-1, were screened out and analyzed based on bioinformatical technology, and hope to explore the mechanism how they involve the interaction between PRRSV and host.Methods:The reliability of the digital gene expression profile (DGE) data was confirmed in three aspects which contain the Sequencing Quality Evaluation, Sequencing Saturation Analysis, Sequencing Saturation Analysis.Differential expression gene associated with interaction between PRRSV and host were screened out in digital gene expression profile (DGE) by bioinformatics analysis.We did some research in gene level and protein level to a better understand on the molecular mechanism of response process of host after infected with PRRSV.Results:The experiments proved that the DGE is reliable and could be used for differential gene screening. NEU1and LAMP-1, which showed down expression in PRRS model’s bone marrow by DGE analysis, found the same results based on QRT-PCR and western blot analysis.Conclusion:neul and lamp-1are two important proteins which involved the exicytosis, pinocytosis, autophagia and immune through lysosome pathway. And the lamp-1is the natural substrate of neul, it can be de-sialylated by the latter. The results of these two genes’ down expression could give us such a conclusion that they involve the PRRSV’s replication and release. But, the details of their mechanism need much more further studies.
