In this exeriment, interspecific hybridation was carried out using Asian lily’Brunello’、’Matrix’and Longiflorum lily ’Formolongi II’、’Raizan II’as materials. Cross-compatibility of different combinations and methods of overcoming barriers were studied. At the same time, pollen tube growth and embryonic development of cross combinations were observed, by means of fluoresence microscopy and paraffin sections, in order to make sure the part of cross barries and provide a basis for overcoming lily cross breeding barries. The main results were as followes:1. Chromosome polidy among4lilies were all diploid:2n=24, except’Brunello’2n=48.2. Pollen germination rate in Longiflorum lily was higher than in Asian lily, which was76.3%in ’Formolongi II’,71.0%in ’Raizan Ⅱ’and64.3%in’Brunello’, The lowest pollen germination rate was60.0%in’Matrix’.3. Murus in pollen surface of Asian lily were consisted with disc and warty beads, while murus in pollen surface of Longiflorum lily were smooth.4. The stigmas of4lilies were triangle、tilobation and medium caity, Asian lily’s stigmas which had monstrosity and trichmata, were smaller than Longiflorum lily’s, but didn’t have exudation, belonging to dry stigma, Longiflorum lily’s which had a few monstrosity、trichmata and exudation, belonging to wet stigma.5. Pollen of cross combination between Asian lily and Longiflorum lily germinated at3-6h after pollination, Pollen tubes passed through the papilla cells of stigma at9-15h after pollination, which began to enter the style channel cells at18h after pollination,and growthed in the style channel cells after pollination21h, following callose reactions, Callose reactions also occurred in stigma and style channel cells, Abnormal pollen tube(tip swelling、tip bifurcation、fold、loopback growth et al.) were emerged both in reciprocal cross,The rate and kind of abnormal pollen tube in reciprocal cross combination was lower than cross combination. In hybridation of’Brunello’and ’Formolongi II’, pollen tube’s longest lenth which’Brunello’as male parent was2.2cm at48h after pollinaton, reaching1/2of the whole style. While a pollen tube of reciprocal cross combination was observed in the ovary at48h after pollination, but not seeing pollen tube tending to the ovule. Reciprocal cross were both belong to prefertilization barries.In hybridation of ’Matrix’and’Raizan Ⅱ’, pollen tube’s longest lenth of cross combinaton which’Matrix’as male parent was2.3cm at144h after pollinaton, reaching 2/3of the whole style, belonging to prefertilization barries, While pollen tube of reciprocal cross combination was observed in the join between style and ovary at144h after pollination, and that the pollen tube had tendency to enter the ovary, futhermore, expand capsule could get at later period and similarly shield embryo were observed at paraffin sections, but not haing any seeds, So we confered reciprocal cross combination was belong to fertilization barries.6. Pollen tube of4self-compatiblitied parents could grow normally and enter the ovary finally, but the time was different. A few’Raizan Ⅱ’’s pollen tubes firstly entered the ovary and tended to the ovule at48h after pollination, The slowest was’Matrix’at96h after pollination, As’Brunello’, at72h after pollination, Pistil of’Formolongi II’entering ovary was not picked, because materials were shortage.According to studies,’Formolongi II "s pollen tube entering ovary was later at48h after pollination.7. Cut style-pollination could oercome cross incompatibility to some degree, and should combine embryo rescue, which just possibly got hybrid progeny.8. Embryos of self-compatibility in Asian lily and Longiflorum lily developed to go though zygotic embryo、proembryo、pear-shaped embryo、shield embryo(clavate embryo)、mature embryo.
