In the study, three techniques, such as the salt solution treatment, the pollen pre-culture and the cut-style method were used to overcome pre-fertilization barriers between'Pollyanna'×'Siberia'('A×O'),'Prato'×'Snow Queen'('A×L') at five flower stages from the bud stage to 3d after anthesis. The results showed that 3.5% KCl and 3.0% NaCl yielded higher seed setting rate (SSR) methods among the three treatments. Then a further research, the effects of 3.5% KCl and 3.0% NaCl on'Pollyanna'stigma morphology and physiological indexes, the paternal pollen tubes growth and the embryo sac and embryo development, were observed and analyzed at five flower stages. It's was expected to investigate the inherent mechanism induced seed setting rate raise and provide references for lily breeding.The main results as follows:1. The effects of KCl and NaCl treatment on SSR, stigma morphology and physiology, pollen germination and pollen tubes growth were similar.2. The effects of salt treatment were better than both pollen pre-culture and cut-style treatment in both combinations. And the treatments of 3.5% KCl and 3.0% NaCl yielded higher seed setting rate.3.'Pollyanna'×'Siberia'got higher SSR than that of'Prato'×'Snow Queen'after salt treatment. Compared with the control, the SSR of the former was raised significantly on 1d-3d after anthesis, and on some treatments it increased 5 to 10 times. Some seeds were gained on the bud stage and bloom day. For the later, it was apt to seed on the bud stage, bloom day and 1 day after anthesis.4. About 90% ovary slices browned in both combinations treatment, and there were enlarged ovules only in 4%-8% slices. The survived seedling rate of embryo rescue significantly increased in'Prato'×'Snow Queen'and it was was better than that of'Pollyanna'×'Siberia'. But there was greater difference among the three sampling times.5. The effects of 3.5% KCl on the morphology of papilla cell observed by scanning electron microscope. All tThe papilla cells of the 5 pollination periods swelled significantly and the viscid exudates covered on papilla cell surface diluted or dissolved, its exposure area increased. Compared to control, the cross and vertical sectional area of papilla cells stages increased (very) significantly at each flowering stage.6. Treated the'Pollyanna'stigma by 3.5% KCl, the activities of the three enzymes were increased by stress in different degree. But the content of soluble protein and resistivity had no significant difference between the control and the treatments.7. The pollen germination percentage improved after treated by 3.5%KCl on the stigma, which caused the quantity of pollen tubes entered style increased and the pollen tubes grew fast.8. Compared with'Prato','Pollyanna'mature embryonic sac peak represent a 3 days lag and its proportion at 8d, 10 d after anthesis was higher, while their double fertilization was observed after 10 d pollination which was later than compatible cross. The embryo sac and embryo degradation and abortion in'Pollyanna'×'Siberia'occurred in 16d to 20d after pollination, while in'Prato'×'Snow Queen'it occurred in 11d to 15d after pollination.9. The optimal inducement condition for embryo seeds germination as follows: at 10℃, 24h darkness treated for 15d. 1/2 MS + 1.0 mg/L IAA and 1/3 MS + 0.3 mg/L NAA were the better media for root of seedlings.
