For the sake of having a complete recognization on heredity foundation of germ plasmresources in semi-wheat and providing scientific reference for parents choice of speciesimprovement in semi-wheat.This paper studied the199kinds of semi-wheat species camefrom HuangHuai area by analysed comprehensively agronomic traits and the genetic diversityat gliadin loci and polymer-phism of molecular markers, the main results were as follows:The analysis comprehensively agronomic traits indicated that the variation scope ofagronomic trait’s Shannon-wiener (Hi) and coefficient of variation (CV) were in1.58-1.99and8.37%-40.87%.the inherit foundation was still narrower than the common wheat inHuangHuai area.The results from gliadin analysis indicated that the average genetic distance among allaccessions was0.403, ranging from0to0.882. This shows that the genetic diversity at gliadinloci of semi-wheat was still lower than the common wheat in HuangHuai area.The result from polymer-phism of molecular markers indicated that14pairs of SSRprimers with stable amplified polymorphic band patterns were screened in21pairs of primers,and a total of45alleles were detected, the molecular weight ranged from127bp to310bp. Thenumbers of different SSR alleles (Na) were quite different, the average was3.21, and thevariation range was2-7. The polymorphism information content (PIC) values ranged from0.049to0.900, the average was0.488. Through the SSR maker cluster analysis, the distanceof4groups of semi-wheat was0and they all come from the same region. Every group hadthe similar agronomic traits, so it indicated that there were some repet between text materials.The analysis to identity rate of cluster and comparison results in gronomic trait, thegenetic diversity at gliadin loci and polymer-phism of molecular markers, there was certainrelationship in three cluster methods, but it was not obvious.In order to select high PHS resistance simi-wheat materials,germination rate (GR) andmolecular marker Vp1B3were used to screen the PHS tolerance of the199semi-wheatmaterials in comparison with two white grain suscep tible cultivars Aikang58and Zhou18.Three types of DNA fragments,845,569and652bp, could be amplified by marker Vp1B3with frequencies of0.05%,34.67%and50.25%in all lines, combined the results of two trials,screened7semi-white wheat KX-11、ZZ-26、ZL-33、AH-57、KQ-65、HY-396、HY-412., they can be used in wheat Pre-harvest sprouting resistance breeding.
