Wheat is one of the important crops. Huang-Huai Region, the main district where yields wheat, portrait in long history, many-cultivars, mature early and high yield, good quality, wide adaptability, contribute greatly to wheat breeding and agriculture development. At present, quality improvement is one of the targets in wheat breeding, and there are considerable ample resources, so it is very important to know the genetic diversity of wheat in this district, to widen the breeding range and to make the breeding and production continual.56 wheat cultivars in Huanghuai were used in current research. First we measured the Kilo-grain weight, Spike per Mu, area of flag leave, length of first stem internodes, length of heading, number of tiller, Spike length and Plant height, and calculate the data to coefficient of variation( CV ) and index of diversity( H '). Then the protein content and SDS sedimentation value of these cultivars were tested, and classified them with K-means cluster, select 10 cultivars by gluten content and biotype. Extracting the genome DNA from the all samples, then we performed amplification using the 10 gDNA samples above as temple and 410 random 10-mer primers. We gained 21 primers amplified polymorphic bands, and used the 21primers to RAPD-PCR, then used the NTSYS-pc2.1 to analysis and got the tree plot. At last, The HMW-GS compositions of wheat cultivars in Huang-Huai region was investigated using SDS-PAGE and the quality scores of each cultivar at Glu-1 have been calculated according to the method described by Payne et al.1. The widely variety of diverse agronomic characters were observed, including Kilo-grain weight, Spike per Mu, area of flag leave, length of first stem internodes, length of heading, number of tiller, Spike length and Plant height. CV were 12.9,15.7,26.76,38.7,28.5,58.2,18.6and8.8 respectively.The variation range of CV was 8.8%~58.2%.The diversity index were 2.09,2.16,2.11,1.90,2.11,2.07,2.10 and 2.11,and variation range of H 'was 1.90~2.16.
