| Tobacco is an important economic crop in China. The cultivated area and yields of flue-curedtobacco in China has become top of the world. Seeds are the basis of tobacco production, and seed vigoris a vital factor for yield. In present days, water and gibberellin solution are mainly used for seedprocessing to improve seed vigor. In this study, to further improve the rate of seed emergence, seedlinguniformity and quality, based on current technologies, we optimize the conditions for seed germination,test the physiological and biochemical indexes for primed seeds, and detect the expression level ofgenes involved in germination. The main results we got are as follows:Fitst, based on the MS nutrient solution and a certain concentration of gibberellins(GA3)for seedpregermination, we additionally add an IAA analogue IBA, it can facilitate seed germination androoting, whose stablity is higher than IAA and security higher than NAA; and add an artificiallysynthesized archusia6-BA, it can promote cell growth, inhibit chlorophyll degradation, improve aminoacid content and delay senescence; The four were mixed in different concentration ratio to process thenew seeds and aged seeds of K326and NC89, respectively. The optimal formula can preferable regulatethe seedling root and stem ratio, and solve the tall seedling issue.Second, MS nutrient solution, GA3, IBA and6-BA were independently used to treat the tobaccoseeds, and all of them can promote germination in proper concentration.1/20MS+100mg/L GA3+2mg/L IBA+0.2mg/L6-BA,1/20MS+100mg/L GA3+2mg/L IBA+2mg/L6-BA and1/20MS+300mg/L GA3+2mg/L IBA+0.2mg/L6-BA, Those optimal formulas are the best ones for seedpregernination. After treatment by these three formulas, the seeds germinate fastly, the germination rateenhanced, the TTC reducing force, POD and NRase activity increased, the conductivity of seed leachliquor reduced, the height of seedlings increased, the dry matter accumulation aggrandized and the seedvigor exceeded CK apparently.Third, According to reported genes on other plants, with RT-PCR technologies, we found one geneWRKY2whose expression level was evidently down-regulated after the pregermination process. TheWRKY2transcription factor mediates seed germination and postgermination arrest of development.Now, there is no research. |
