Objective:This study is based on production practice and the researches of some representative dairy farms in north Xinjiang, mainly focusing on the problem of a high incidence of infertility It applies rectal palpationand B-mode ultrasound to make a comprehensive diagnosis on cows with wovarian disease infertility, adopting optimize hormone programs, orally traditional Chinese medicine Cyst-regulating Power and Fertility-promoting Power respectively to compare its treatment effect. By observing the of morphological changes of ovarian and uterine before and after the application of B-mode ultrasound, a comparatively complete B ultrasound image of ovarian disease can be summarized before and after the treatment, which is of great referential value and directive significance for future research and production practice. With the rules of variation of P4, E2, LH, and FSH in the serum of sick cows before and after the traditional Chinese medicine, it provides relevant theories on further exploration in pharmacological sac scattered, progestational casual treatment of ovarian disease, infertility and the mechanisms in the future.Testing methods and results:1. Investigation and analysis of the common ovarian disease infertility on large-scale dairy farms.From the materials of some representative cattle breeding in Kuitun and Shihezi from2008to2012(calving, breeding record, age, parity, body condition, empty cup days, and etc), combined with the clinical symptom, surveys on the the incidence of infertility of these cows in the above region have been carried out at random. The results showed that infertile cows with ovarian disease (ovarian cysts, delayed ovulation, ovarian inactivity, ovarian of persistent corpus luteum) are the most common, followed by uterine disease. The incidence of infertility of ovarian disease is closely related to parity, body condition, season and other factors.2. Diagnosis on the common ovarian disease of cows.By the application of rectal palpation and B-mode ultrasound combined with hormone determination,233cows are diagnosed with ovarian disease on the basis of the exclusion of uterine diseases and other diseases, including66cows with ovarian cysts,38with delayed ovulation,91with ovarian inactivity, and38with ovarian of persistent corpus luteum.3. Researches of the application of optimization the hormone to treat cows with common ovarian diseaseConventional hormone therapy program combined with the injection of VADE and miscarriage measures different stages after mating is applied to treat ovarian cysts, delayed ovulation, ovarian inactivity, and ovarian of persistent corpus luteum,20for each. Surveys are carried out on the cows before and after the application of B-mode ultrasound. The results showed the cure rates of the four diseases were60%, 55%,55%,70%, higher than that of conventional hormone. The change in B-mode ultrasound image is obvious before and after the treatment.4. The study of dynamic monitoring of B-mode ultrasound on common ovarian disease in dairy cows before and after Taking orally traditional Chinese medicine treatment15dairy cows of Ovarian cysts and delayed ovulation take orally Cyst-regulating Power;15dairy cows of ovarian inactivity and persistent corpus luteum take orally Fertility-promoting Power. After1-2courses, B-mode ultrasound dynamic follow-up examination is applied. The results showed that the cure rates of ovarian cysts, delayed ovulation, ovarian inactivity and persistent corpus luteum were66.7%,60%,60%,60%, higher than the optimization the hormone program (except persistent corpus luteum). After taking orally medicine the change of length of cyst ovaryas well as the width of cyst ovary was not significant (P>0.05). The longitudinal diameter of horn uterus declined significantly on the16th day.(P<0.05); The change of longitudinal diameter of cervix uterus was not significant(P<0.05),but the size of horn uterus was smaller than that before the treatment. The changes of delayed ovulation morphological indexs of ovaries before and after taking medicine were not significant (P>0.05), but all reached normal ovarian size. The change differences of longitudinal diameter of horn uterus were significant after finishing treatment on the7th day, the10th day and the13th day (P<0.05); The change differences of longitudinal diameter of cervix were not significant (P>0.05).The length of ovary of cows with ovarian inactivity increased significantly (P<0.05) after finishing treatment on the1st day, the4th day, the10th days and the19th day, compared to that of dairy cows before taking medicine. The longitudinal diameter of cervix uterus increased significant after finishing treatment on the13th day and the16th day (P<0.01)and on the19th day (P<0.05). The longitudinal diameter of horn uterus was not significant(P>0.05). Medium and small follicules of persistent corpus luteum in dairy cows appeared in ovary after finishing treatment for1day and most of large follicules appeared after finishing treatment on the10th day and the19th day respectively and part of them appeared estrus and ovulation. The length of ovary on the right and left side did not decrease significantly (P>0.05) after finishing treatment compared to that of dairy cows before taking medicine. The longitudinal diameter of cervix uterus and horn uterus increased not significant after finishing treatment (P>0.05)-5.The change of hormone concentration of P4, E2, LH, FSH in ovarian disease cows before and after taking orally Cyst-regulating Power and Fertility-promoting PowerThe application of ELISA method to measuring the level of P4, E2, LH, FSH in serum of cows in normal group and control group showed that levels of E2, P4of cows with ovarian cysts in control group were higher than the level of the normal group, and that the levels of LH, FSH of ovarian cysts in control group is lower than that of the normal group; After taking orally Cyst-regulating Power,the level of E2, LH, FSH in serum of cows increased, and the level of P4declined. The level of E2of ovarian cysts in cows increased significantly after taking medicine on the7th day (P<0.05). The level of LH and FSH increased significantly after taking medicine on the10th day and the19th day (P<0.05). the level of P4didn’t decline significantly (P>0.05). For the cows with delayed ovulation,the level of P4was lower than that before taking medicine (P<0.01) on the13th, the16th, and the19th day. the level of E2was higher than that before taking medicine (P<0.05) on the13th, the16th, and the19th day. the level of LH was higher in7,13days than that before taking medicine(P<0.05) on the7th and the13th day. And on the16th and the19th days was extremely higher than before taking medicine (P<0.01); the level of FSH was higher on the10th day than before taking medicine(P<0.05). For the cows with ovarian inactivity, the level of P4, E2, LH, FSH was higher than before taking medicine, but not significant (P>0.05). For the cows with persistent corpus luteum, the level of E2and FSH was higher than before taking medicine, especially on the10th, the13th, the16th, and the19th day (P<0.05) and the level of FSH increased but not significant(P<0.05). The level of LH was declined not significantly after taking medicine (P<0.05). The level of P4declined more significantly on the16th and the19thday than before taking medicine(P<0.01).
