Natural populations of Pinus koraiensis in Fenglin national natual reserve, Yichun city, Heilongjiang Province; Hongwei forst farm, Lushuihe Foresty Bureau, Jilin Province; Baishilazi national natual reserve, Kuandian county, Liaoning Province were the sample sites in this paper. The mating systems and the genetic diversity of49individuals were studied by cpSSR marker methods.The392offsprings of natural populations of Pinus koraiensis were amplified by five cpSSR primers.18loci were amplified, including11polymorphic loci. The number of Polymorphic bands was ranged from2to3. The average number of polymorphic loci was2.2. The primer69F/R had3polymorphic loci and the other primers had2polymorphic loci. The length of microsatellite fragments was ranged from130bp to500bp.The mating systems of49individuals were analyzed that single-locus(ts) outcrossing rate was0.756(SE=0.071), multi-locus(tm) outcrossing rate was0.790(SE=0.073), imbreeding index(tm-ts) was0.034(SE=0.023) that was closed to0. Accoding the result of the imbreeding index, there was no imbreeding recession among natural populition. The constant index(F) was-0.057(SE=0.000) that was below0. Accoding the result of the constant index, there was excessive heterozygote.The mating systems of3different areas were analyzed. In Fenglin, multi-locus(tm) outcrossing rate was0.962(SE=0.000), single-locus(ts) outcrossing rate was1.116(SE=0.000), imbreeding index(tm-ts) was-0.154(SE=0.000), the constant index(F) was-0.300. In Lushuihe, multi-locus(tm) outcrossing rate was0.804(SE=0.000), single-locus(ts) outcrossing rate was0.778(SE=0.000), imbreeding index(tm-ts) was0.026(SE=0.000), the constant index(F) was-0.300. In Baishilazi, multi-locus(tm) outcrossing rate was0.767(SE=0.000), single-locus(ts) outcrossing rate was0.771(SE=0.000), imbreeding index(tm-ts) was-0.004(SE=0.000), the constant index(F) was-0.077. The imbreeding index of each area was closed to0, showing no imbreeding recession among natural populition. The constant index of each area was below0, having excessive heterozygote in natural populition. The results show that the order of multi-locus and single-locus outcrossing rate in3different areas was Fenglin, Lushui, Baishilazi. Analysing between the mating systems and the geographic location, multi-locus and single-locus outcrossing rate rise as the latitude and longitude rise. Analysing between the mating systems and the distribution area, the multi-locus and single-locus outcrossing rate of the center was higher than the edge. Analysing between the mating systems and the population characteristics, the multi-locus and single-locus outcrossing rate of the over-mature forest was higher than the mature forest, the multi-locus and single-locus outcrossing rate of old forest was higher than young forest, the multi-locus and single-locus outcrossing rate reduce as the population density rise.The genetic diversity of3different areas was analyzed. In Fenglin, the effective number of alleles was1.1058, Shannon diversity index was0.1481, Nei diversity index was0.0835. In Lushuihe, the effective number of alleles was1.3009, Shannon diversity index was0.2568, Nei diversity index was0.1727. In Baishilazi, the effective number of alleles was1.1887, Shannon diversity index was0.1963, Nei diversity index was0.1241. The results show that the order of the genetic diversity in3different areas was Lushui, Baishilazi, Fenglin. Analysing between the mating systems and the genetic diversity, Both of chang rules were different.The gene differentiation of3different areas was analyzed. The percentage of genetic diversity within Korean pine population was87.46%in the total value, The percentage of genetic diversity be among Korean pine populations was only12.54%, that means the genetic variation is mainly occurred inner Korean pine population.
