Insulin-like growth factor-1was a kind of multifunctional bioactivepeptides,which had a direct impact on animal growth, development, reproduction,nutrition, metabolism and tissue cell proliferation, differentiation, apoptosis andcancer,meanwhile regulated the animal life activities,but was just one of severalpolypeptide growth factors. Neuropeptide Y involved in the control of food intake,regulation of hormone release, sexual behavior, metabolism, blood pressure, circadianrhythms, and reproduction, and so on,and study also found that NPY was an importantsatiety factor, which had important significance to improve the quality oflivestock.Wuding chicken were took for experiment subjects.Using PCR-RFLPmethod investigated the correlation between candidate genes NPY gene, IGF-1genepolymorphism and Wuding chicken laying performance, which aimed at protectingbiodiversity of animal genetic resources, making full use of and cultivating to meetthe needs of the community to provide a sound scientific basis. The experimentalresults were as follows:1PCR-RFLP analysis showed that wuding chicken NPY gene TSS area and exon2existed Dra I and Psp1406Ⅰ restriction enzyme sites,respectively. There lacked offour bases in the Dra Ⅰ bit point;Psp1406I point exists A→G mutation and thepolymorphic loci was never been reported. IGF-1gene UTR region of Hinf Ⅰrestriction site had the existence of the point C→T mutation.2Through analyzing NPY gene and IGF-1gene polymorphic fragments,we detectedgene and genotype frequencies statistics and found that Dra Ⅰrestriction enzymelocus allele A frequency was0.7, the allele B frequency was0.3; Psp1406I pointallele C frequency was0.16,the allele D frequency was0.84; Hinf I sites allele Efrequency was0.53, the allele F frequency was0.47. By chi-square test, three restriction sites of Wuding chicken two genes were in Hardy-Weinberg equilibrium.3Using NPY gene genotype effects least squares analysis found that NPY (TSS) geneDra I restriction enzyme site polymorphic was related with Wuding chicken eggopening,240-day-old egg number and egg weight and39-week-old egg number andegg weight. AB genotype individuals open egg weight were significantly higher thanAA and BB genotype individuals(P <0.01). AA and AB genotype individuals weresignificantly lower than BB genotype individuals in the240-day-old egg number and39-week-old egg number and egg weight(P <0.01), AA and AB genotype individualswere significantly lower than BB genotype individuals in the240-day-old eggweight(P <0.05). NPY (exon2)gene Psp1406I restriction enzyme site polymorphicwas related with Wuding chicken the open egg weight, egg number of39-week-old.DD genotype individuals open egg weight were significantly lower than the CDgenotype individuals(P <0.01), CD genotype individuals were significantly lower thanthe DD genotype individuals in the39-week-old egg number(P <0.05).4Using IGF-1gene genotype effect least squares analysis found that IGF-1(UTR)gene Hinf Ⅰ restriction enzyme site polymorphic was related with Wuding chickenegg open,240-day-old egg number and egg weight and39-week-old egg number andegg weight. EE genotype individuals open egg weight were significantly higher thanthe EF and FF genotype individuals (P <0.05), EE and FF genotype individuals weresignificantly higher than EF genotype individuals in the240-day-old egg number andegg weight in the39-week-old egg number(P <0.01), EE genotype individuals weresignificantly higher than the EF genotype individuals(P <0.01), and there weresignificant differences between EE genotype individuals and the FF genotypeindividuals(P <0.05).
