Metabonomics techniques were used to analysis Angelica on plasma, spleen,liver tissueand urinary metabolites change in blood deficiency mice caused by APH and CTX, from theperspective of changes in metabolites studied the protective mechanism of Angelica on defi-ciency of blood mice. Kunming mice were randomly divided into blank group, blood defi-ciency model and Angelica decoction liquid high dose group (20g/kg), Angelica decoctionliquid middle dosage group (10g/kg) and Angelica decoction liquid low dose group (5g/kg),tested different groups of Red Blood Cells (RBC), White Blood Cells (WBC), Hemoglobin(HGB), Platelet (PLT) and thymus and spleen index. Using GC-MS analyzed different groupsof metabolites, and the orthogonal partial least squares (OPLS-DA) and principal componentanalysis (PCA) analyzed the different groups of variation of the metabolites in the plasma,spleen tissue, liver tissue and urine and looked for markers and results were as follows.1. Study of Angelica decoction liquid on the blood-deficient model mice by APH andCTX result: compared with blank group, RBC, WBC, HGB, PLT content and in thymus in-dex, spleen index significantly reduced in the model group (p<0.01). Compared with modelgroup, Angelica decoction liquid different dose groups had different of degrees increased inthe peripheral blood of RBC, WBC, HGB, PLT content and the thymus index, spleen index,among Angelica decoction middle dose group worked best. Angelica could improve RBC,WBC, HGB, PLT content and the thymus index, spleen index of blood deficiency mice,which was account for Angelica had enriched blood.2. Angelica on blood deficiency model mice of plasma GC-MS tested result:24kinds ofmetabolites were identified in the plasma of different groups, mainly including the fatty acid,sugars and amino acids. Sample points of blank group were separation from sample points ofmodel group in the PCA score matrix diagram, which explained that the normal physiologicalfunction of molding mice were disturbed, from the body’s physiological endogenous metabo-lites change levels could also think blood deficiency model building success. VIP (variableimportance projection)method was used to find the different biomarkers between everygroup.Compared with blank group, model group of Lactic acid, Arachidonic acid, Stearic ac-id, Glycine Malic acid, Palmitic acid, Threonine, Valine of content increased and Proline of content decreased (p<0.05); Compared with model group, Angelica decoction liquid differentdose groups of Lactic acid, Arachidonic acid, Stearic acid, Glycine Malic acid, Palmitic acid,Threonine, Valine of content decreased and Proline of content increased (p<0.05). Comparedwith blank group, these substances change close to the blank group, Angelia decoction liquidmiddle dose group effect was best.3. Angelica on blood deficiency model mice of spleen tissue GC-MS tested result:21kinds of metabolites were identified in the spleen tissue of each group’s, mainly including thefatty acid, sugars and amino acids. Sample points of blank group were separation from sam-ple points of model group in the PCA score matrix diagram. VIP method was used to find thedifferent biomarkers between every group.Compared with blank group, model group of Lac-tic acid, Stearic acid, Malic acid, Butyric acid, Arachidonic acid, Threonine, Valine, Glycineof content increased and Alanine, Proline and Fructose of content were decreased (p<0.05);Compared with model group, Angelica decoction liquid different dose groups of Lactic acid,Stearic acid, Malic acid, Butyric acid, Arachidonic acid, Threonine, Valine, Glycine of con-tent decreased and Alanine, Proline and Fructose of content were increased (p<0.05). Com-pared with blank group, these substances change close to the blank group, Angelia decoctionliquid middle dose group effect was best.4. Angelica on blood deficiency model mice of hepatic tissue GC-MS tested result:21kinds of metabolites were identified in the hepatic tissue of each group’s, mainly includingthe fatty acid, sugars and amino acids. Sample points of blank group were separation fromsample points of model group in the PCA score matrix diagram. VIP method was used to findthe different biomarkers between every group.Compared with blank group, model group ofLactic acid, Alanine, Valine, Stearic acid, Glycine, Palmitic acid, Threonine, Butyric acid,Malic acid of content increased and Fructose of content were decreased (p<0.05); Comparedwith model group, Angelica decoction liquid different dose groups of Lactic acid, Alanine,Valine, Stearic acid, Glycine, Palmitic acid, Threonine, Butyric acid, Malic acid of contentdecreased and Fructose of content were increased (p<0.05). Compared with blank group,these substances change close to the blank group, Angelia decoction liquid middle dose groupeffect was best. 5. Angelica on blood deficiency model mice of urine GC-MS tested result:21kinds ofmetabolites were identified in the urine of each group’s, mainly including the fatty acid, su-gars and amino acids. Model mice of urine metabolites by APH and CTX with time trajectoryanalysis found that in9d, model group mice of endogenous metabolites suffered the the big-gest influence in urine. VIP method was used to find the different biomarkers between everygroup.Compared with blank group, model group of Glucose content increased and Citric acid,Succinic acid, Glycerol, α-ketone glutaric acid and Xylitol of content decreased (p<0.05);Compared with model group, Angelica decoction liquid different dose groups of Glucosecontent decreased and Citric acid, Succinic acid, Glycerol, α-ketone glutaric acid and Xylitolof content increased (p<0.05). Compared with blank group, these substances change close tothe blank group.Angelica could alleviate blood deficiency symptoms in mice by APH and CTX, characte-rized by RBC, WBC, HGB, PLT levels and immune organ thymus index, spleen index wereincreased. Applyed GC-MS method to detect plasma、spleen tissue、liver tissue and urine ofmetabolite changes in mice, mainly involved the hinder flocculant fatty acid metabolism,energy metabolism and amino acid metabolism disorder. Fat metabolism disorders, cells in-side and outside osmotic pressure damage and loss of glycolysis function was the mechanismof blood deficiency syndrome caused by CTX and APH. Angelica blood tonic action mechan-ism was related to improve mitochondrial function and energy metabolism, lipid metabolism,maintaining cell inside and outside osmotic balance and improved immune function.
