Liquorice is the important sand binding plant in the west desert and semi-desert area of China.Before the1980’s, finished product of liquorice mainly depends on excavation of wild liquorice,which cases water and soil loss and desertification. Liquorice seeds for artificial cultivation are sofar mainly from the wild, which varietal complexity and the seeds are scarce. And germination rateof wild liquorice seeds are very low. Now wild liquorice seeds can not satisfy the requirement ofproducing, which became the biggest bottleneck of licorice industry development. The study usethe tissue culture technique which take the axillary bud and stem tip of licorice that no vaccine andthe root, stem and blade of2years licorice as the explant to study their optimum medium andhormone. The aim of the study is to build three intermediate propagation systems of liquorice,which lays a solid foundation for solving the problem of liquorice producing. The mainconclusions are as follows:1. The results indicated that the best culture medium was MS, the most suitable Lengths ofStem Segments was0.7cm, the best concentration was1mg/L6-BA, the most suitable cultivationcondition was MS+6-BA1mg/L+NAA0.1mg/L, lengths of Stem Segments was0.7cm, culture firstgeneration need20days, increment multiple was7, one Radix Glycyrrhiza seed can become117649aseptic seeding after half-year of cultivation.2.the material of Glycyrrhiza uralensis Fisch shoot-tips. Results showed that culturemediums had the greatest impact,6-BA was the minor, Sucrose had the least impact. Thesuitable hormone proportion of culture medium for Glycyrrhiza shoot-tip was MS+6-BA0.3mg/L+NAA1.0mg/L+KT0.3mg/L+sucrose30g/L. The suitable consists of proliferation medium was MS+6-BA1.0mg/L+NAA0.1mg/L+sucrose30g/L, proliferation ratio reached7times,average plant height was5.4cm; The suitable consists of rooting medium was MS+KH2PO413.5mg/L+sucrose30g/L and rooting rate reached100%.3. it is difficult that Glycyrrhiza uralensis Fisch blade induce cluster bud, but it canproduce callus; the induction rate of root tip which induce the cluster bud is low; the induction rateof stem tip induced by cluster bud is the highest; The best medium consist is: stem tip+KT0.01mg/L+NAA0.5mg/L+and sucrose.30g/L The best propagation medium combination is:1/2MS+KT0.5mg/L+NAA0.5mg/L+sucrose30g/L, it Takes much more root number, root length is longer, the growth of plant is thrive.
