Experiments were conducted to study the effects of nanomaterialsRhodiola Sachanensis Polysaccharide on cryopreservation of boar semen.The results were as follows:1.In our study,Rhodiola Sachalin ensis Polysaccaride (RSP) andnanomaterials Rhodiola Sachalin ensis Polysaccaride(NRSP) were addedinto the basic extenders. After frozen-thaw,the motility of sperm, theintegrity rate of sperm acrosome and other quality of cryopreservation ofboar semen were investigated. Moreover,MDA quantity and the activity ofSOD were detected by the related kit. The results showed that when theaddition of NRSP was15%(V:V), there were significant differencesbetween experiment and control groups in motility of boar sperm, theacrosome integrigy and the membrane integrity of boar sperm afterfreezing and thawing(P<0.05), and they were0.69,51.42%,25.95%,respectively. Extender containing15%NRSP enabled the percentage of thefirst class sperm(P<0.05).With the addition of NRSP in the dilution offrozen semen, the MDA quantity was decreased. When containing15%NRSP or6mg/l RSP,the MDA quantity was lowest(P<0.05). And theactivity of SOD increased as the addition of NRSP in the extender more up. The result of present study indicated that there was a significant positivecorrelation among sperm motility, membrane integrity and acrosomeintegrity(P<0.05). MDA quantity has a significant opposite correlationwith sperm motility, acrosome integrity(P<0.05);2.In our study, we combine15%NRSP and different concentrationegg yolk adding into basic extenders.The result shows that when adding afinal concentration of3%of glycerol, sperm motility, plasma membrane,acrosome intact rate integrity rate, living rate were significantly higher thanthat without glycerin.When the final concentration of2%group the effect isobviously better than without glycerin added group (P <0.05); Adding15%NRSP with different concentration of yolk compatibility, shows that whenthe concentration of yolk is20%and30%, the semen quality is best. Thesperm motility, plasma membrane, acrosome intact rate is significantlyhigher than10%concentration of yolk add group (P <0.05), but the groupof20%and30%yolk concentration has no significant difference in spermacrosome intact rate and plasma membrane integrity rate (P>0.05);3.0.5ml straws were respectively placed at the height of1,3,5,7and9cm above liquid nitrogen for12min in polyethylene box.After freezing–thawing detect sperm motility, plasma membrane, acrosome intact rate ofboar semen and the motion state, the results found that after freezing-thawing the3cm group sperm motility, plasma membrane, acrosome intact rate is the highest, and significantly higher than other frozen height (P <0.05). Besides1cm and9cm height frozen group the sperm motility islower than0.3cm, which is not suitable for used in pig semencryopreservation. The sperm of type A of3cm was the highestafter thawing;Sperm pre-equilibrated at17℃temperature and at the normaltemperature in the process of sperm freezing-thawing activity there wasno significant difference in membrane structure (P>0.05); there were nosignificant difference found between each step of process of filtering,17℃after balance、add refrigerant Ⅰextender,before freezing,after thawed(P<0.05);4.Thisstudy was performed to optimize freezing and thawing protocolof rapid boar seminal cryopreservation.To compare the different thawingtemperature and thawing time affect on sperm quality, the results showedthat52℃,9sgroup’s sperm quality was the best, sperm motility, plasmamembrane, acrosome intact rate was significantly higher than others(P <0.05), and the sperm quality differences between different thawing timesignificantly (P <0.05); Adding different concentration of RSP and NRSPin the BTS, the group of15%NRSP and2mg/L RSP added vigor is best,but havn’t the obvious difference (P>0.05), the15%NRSP group thawedsperm acrosome intact rate and plasma membrane integrity ratesignificantly better than other groups (P <0.05);15%NRSP and yolk compatibility, thawed sperm compared with single NRSP group qualitydrops, and significant difference (P <0.05).
