Pollens carry out the crucial functions during the process of plant sexual reproduction.What’s more, the developments of pollens are the very complex processes because thereare so many genes involved in those processes. Studies on the pollen specific promoterswill help researchers to better understand the molecular basis of pollens’ developmentsWheat is one of the most important crops all over the world and due to its huge andcomplex genome, the research progress regarding wheat is behind other important cropslike rice on molecular level. The researches on wheat pollen specific promoters haveessential help for the understanding of the molecular basis of wheat pollens’ developmentand also provide useful and important information to build male sterility for hybrid seedproduction.In this study, the promoters of PSG076and TaPSG076genes were analyzed usingbioinformatics and transformation methods. The1.4kb and1.7kb promoter fragments ofPSG076and TaPSG719genes were amplified respectively and the P14G and the P17Gvectors, which contain the GUS gene and promoter fragments (1.4kb and1.7kb), wereconstructed. P14G and P17G vectors were introduced into wheat immature embryos byusing particle bombardment technique. Regenerated wheat plants were obtained aftertissue culture period of bombarded wheat immature embryos. PCR technique confirmedthat the vectors were inserted into the wheat genome. Histochemical assays of GUSactivity results showed that the1.4kb promoter fragment of PSG076gene is the pollenspecific promoter and it does effect during the late pollen developmental stages.
