The subject for issues of abnormal wound roots, long seedling cycle, and low survival rateexisting in Dendrobium (Dendrobium officinale Kimura et Migo) tissue culture production, carriedout root control technology, growth and accumulation mechanism of effective components andOne-step Culture study for Dendrobium officinale tissue culture seedling, to optimize root controltechnology from the concentration of banana juice, root cutting treatment and nitrogen ratio, andprove growth and accumulation mechanism of effective components for Dendrobium officinale, toexplore One-step Culture for Dendrobium officinale by use of tissue culture and soillesscultivation method, provide the basis for industrial production. The results showed that:1The study of root control technology showed that: Banana’ concentration of70g/L was thebest for root control; Root cutting treatment had significant effect on root control, but affectedplant growth to some extent.;(NH4+—N):(NO3-—N)=40:20in MS medium was the best for rootcontrol.2The study of accumulation mechanism on effective component showed that: Bananas’concentration of90g/L was conducive to accumulation of effective components, of whichpolysaccharide production reached3.7368×10-2g and alkaloid production reached2.9×10-5g;Polysaccharides production of first-generation and second-generation seedlings after root cuttingwere significantly higher than those of no root cutting; contrast to third generation seedlings.Alkaloid production of first-generation seedlings after root cutting had no significance to those ofno root cutting; alkaloid production of second-generation and third generation seedlings after rootcutting were significantly lower than those of no root cutting; Polysaccharide production of (NH+4—N):(NO3-—N)=60:0in MS medium was the best, reaching4.2323×10-2g. Alkaloid productionof (NH+4—N):(NO-3—N)=50:10was the best, reaching1.0×10-5g; Polysaccharide production ofratio3:1of banana and banana’s peel of was the best, reaching1.5814×10-2g; alkaloid productionof ratio0:1of banana and banana’s peel was the best, reaching1.9×10-5g; light intensity of2000Lx had the best effect on the accumulation of effective components, of which polysaccharideand alkaloid production reached4.5328×10-2g and1.7×10-5g; Polysaccharide production ofdouble-hole lid was the best, reaching2.2325×10-2g; alkaloid production of three-hole lidreached maximum1.4×10-5g. 3One-step Culture set tissue culture and soilless cultivation method in one, with a shorttraining period, simple process, low cost, high transplanting survival rate. In the plug liquidculture process, fresh weight, dry weight and survival rates of Dendrobium grew in the nutrientsolution1/4MS were better, respectively,2.35g,0.19g and92.86%, better than other treatments.
