In the paper, Robinia pseudoacaciaf.‘Unifolia’ and Agapanthus praecox ssp. rientalis‘Big Blue’ were used as materials to established the system of tissue culture and rapidpropagation. A series of research about light curve and cold resistance of R.‘Unifolia’ hadbeen studied. The results showed as follows:1. Established the rapid micropropagation system of R.‘Unifolia’. The best initialmedium was: MS+6-BA1.0mg/L+NAA0.2mg/L and the sprouting rate85.67%. The bestproliferation medium was: MS+6-BA1.3mg/L+NAA0.2mg/L, in which the highestproliferation coefficient was4.88. Macroelemen affected the rooting the rate significantly,when they were1/2of MS, the average rooting rate of R.‘Unifolia’ a reached its highest of57.45%, and the average root number reached3.15. The optimal rooting medium was1/2MS+IBA0.5mg/L+NAA0.3mg/L. Leaves were more suitable for callus induction thantender shoots. When the medium with concentration of6-BA6.0mg/L and2,4-D2.0mg/L,the highest rate of callus induction was60%. The best differentiation rate(74.1%) ofadventitious bud form callus was obtained on the followed medium: MS+6-BA1.5mg/L+NAA0.2mg/L.2. Established the rapid micropropagation system of A praecox ssp. rientalis ‘BigBlue’. The optimum medium for rhizome was MS medium containing6-BA4.0mg/L+NAA0.1mg/L. The shoot propagation medium: MS+6-BA4.0mg/L+NAA0.1mg/L. Onthe medium1/2MS+IAA0.5mg/L, the elongated shoots rooted at the rate of80%.3. Through the analysis of the data of the grafting experiments with completerandomized block design of concentration of hormone, kinds of hormone, coarse of rootstock, the result showed that NAA and IBA+NAA were better than IBA. The bestcombination was as followed: the coarse of stock between5.5~7.5cm and NAA200ppm.4. Photosynthetic charateristics R.‘Unifolia’ was measured by using Li-6400potablephotosynthesis system. The curve of diurnal variation of R.‘Unifolia’ showed two peaks.The concerntration of intercellular CO2presented a valley state at lunchtime from12:00to14:00. The diurnal variation of intercellular CO2demonstrated the contrary regularity tothat of the photosynthetic rate, transpiration rate, and stomatal conductance. In thephotosynthetic monthly varistion, the photosynthetic rate of R.‘Unifolia’ showed anexcellent performance on the upswing. 5. In early frost hardening, the cold resistance of R. unifolia was higher than R.‘Linn’,which was similar to R.‘decaisneana’, R.‘Tortuose’. From December to January of thefllowing year, the cold resistance of R.‘Unifolia’ was significantly higher than the otherthree species. During frost hardening, the soluble sugar content of R.‘Unifolia’ was higherthan that of other three species while the starch content was lower than others’. As theresult, the cold resistance of R.‘Unifolia’ was higher than R.‘decaisneana’、R.‘Linn’、R.‘Tortuose’.
