The aim of present study was to Detect SNPs intron2and exon3,3’UTR in adiponectingene (ADPN) and5’ UTR and exon2and intron2insulin-like growth factor1(IGF1) in smalltail han sheep population containing259individuals, by combining DNA mixed poolsequencing technology with direct sequencing. Distinction between homozygous andheterozygous of The sequencing results were distincted by condoncode4.0program; Geneticpolymorphisms of the gene mutation sites were analyzed by Popgen3.2and Molkin3.0Software; Gene linkage disequilibrium analyzed by SHEsis Software; the Correlation analysisof polymorphisms with little size traits, Body Size Body Type Index and Blood physicaland chemical properties were Estimated by SAS9.0program. Results as follows:(1)Six SNPs have been Detected, G/Aat10436bp, A/G at10454bp、T/C at10660bp,C/T at10677bp、C/T at10732bp and C/T at10753bp in intron2and exon3in ADPN gene.The distribution of alleles and genotypes were under Hardy-Weinberg equilibrium. The10436and10660site,10677and10753were in complete linkage, through Linkagedisequilibrium, Respectively. Two SNPs have been Detected, C/T at4908bp、G/A at4988bpin intron2and exon2in IGF1gene, and all under Hardy-Weinberg equilibrium.(2)The difference between genotypes in10454site was significant for The FourthLitter size at significant level(P<0.05)in ADPN gene. However, the Third Litter size trait ofthe CT genotype was highly significantly (P<0.01) higher than CC genotype, and There was asignificant allele substitution effect (P <0.05), yet The allele A was Favorable alleles ofregulated Litter size in4988site.(3)There was a difference of traits of each locus inADPN gene in different age, but notin Genotype and Interaction effects. However, Interaction effects in4988site is significantly(P <0.05) with HH and BH in IGF1gene, Specifically, The difference between genotypes in2year-old was significant for HH and BH at significant(P<0.05) and highly significantlevel(P<0.01). The allele A had negative effect on body length index in1-year-old in4988site.(4)The difference between genotypes at each locus was significant for ALP at significant level(P<0.05) or highly significant level(P<0.01) in different age or all population.10732site is highly significantly (P <0.01) with TG in ADPN gene in5-year-old age and allpopulation.4908site is significantly (P <0.05) highly significant level(P<0.01) withALP,CHO and HGB in IGF1gene in different age and all population.(5) Interaction effects and Haplotype in ADPN gene is significantly (P <0.05) or highlysignificant (P<0.01) withPhysiological indicators and Biochemical indicators; Interactioneffects and Haplotype in IGF1gene is significantly (P <0.05) or highly significant (P<0.01)withPhysiological indicators and Biochemical indicators;(6) Interaction effects is significantly (P <0.05) or highly significant (P<0.01) with Littersize, BH,Physiological indicators and Biochemical indicators between ADPN and IGF1gene.This study found that ADPN, IGF1gene is related to Litter size, Physiological indicatorsand Biochemical indicators, Specifically, the mutation at some locus have significant effect ontraits that can be used as molecular genetic markers for part of the production traits of smalltailed han sheep; There are Interaction between ADPN and IGF1genes, that can can build theadvantage composite genotype in order to accelerate the breeding process.
