Jining Grey goats, widely distributing in Jining and Heze regions in Shandong Provinceof China, are noted for the wavy patterns of its kid-pelt which is the traditional commodity ininternational markets and display significant performances of sexual precocity. The JiningGrey goats early sexual maturity, highly reproduction rate, fetal and other biologicalperformance reasons mechanism is at home and abroad for many years researchers payattention to scientific questions, whether with GHR、IGF-IR expression, distribution andfunction have inherent contact it is not entirely clear. At present, Jining Grey goats whichGHR、IGF-IRgene has not been reported.The objective of the experiment was to study the localization of GHR and IGF-IR andthe expression of GHR mRNA and IGF-IR mRNA in uterus of Jining Gray Goat. WesternBlot: detect the protein fragment size of GHR, IGF-IR. RT-PCR: detect and analyze theexpression regularity of GHR mRNA and IGF-IR mRNA.The immunohistochemical staining(SABC):detect the location of positive cell of GHR and IGF-IR.The results showed that:Western Blot measure specificity protein fragments of GAPDH, GHR, and IGF-IR at36KDa,70KDa,55KDa.GHR and IGF-IR had similar variations,GHR and IGF-IR plays an importantrole in the growth of the uterus, also at the beginning of the sentiment time and the naturalmature period are the womb grow the quickest time, enable the womb physiology function totend to the consummation, provides the nutrition for the embryo bed and the early embryo toprepare for.mmunoprotective staining was mainly localized in the cytoplasm of glandularepithelial, stromal cells, vascular endothelial and uterine smooth muscle cells, occasionallysaw the nucleus coloration, and GHR masculine gender dyeing number is more than IGF-IR.The expression of IGF-IR mRNA and GHR mRNA and their positive cells have a samevariation tendency, but the expression quantity of IGF-IR mRNA is less than GHR mRNA.The expression quantity of IGF-IR and GHR have reached a top in the60th day and the120thday(P<0.01), then remained at a relatively stable level after the fall. Conclusion: GHR andIGF-IR play an important role in the growth of the uterus, and the puberty and the sexual maturity may be the fastest period of the uterus during postnatal development, enable thewomb physiology function to tend to the consummation, provides the nutrition for the embryobed and the early embryo to prepare for.
