TBC1D1is a Rab-GTPase-acting related protein implicatied in regulating the trafficking of glucose transporter4(GULT4) in response to cell signal transduction and material transport,and it play an important role in sugar metabolism and fat metabolism. Currently,the carecass performance and its improvement is a major issue in poultry breeding, and it can be used as one of the most important basis for measuring production performance. How to research its related traits in molecular level that in order to find some proper genetic markers has become an important direction of poultry breeding. Then, many researches on human, mouse and the procine showed that it could be a appropriate candidate gene for human obesity or livestock and pourlty carcass traits.The polymorphisms of TBC1D1gene were detected by molecular biology method among64chickens from two populations(including the pure lines of SD02and SD03in the first generation of Erlang Mountainous Chicken).On the basis of the data, the study of the assciation with some carcass traits, such as live weight, body weight,semi-eviscerated weight, eviscerated weight, breast muscle weight, thigh weight and abdominal fat weight was carried out.It may provide a theoretical basis for further research.The results were as follows:There were17SNPs were dectected by direct sequencing in target region, which located in the region of seven exons and part of three introns,and most of them were synonymous mutations. There were six SNPs located in three introns while eleven SNPs located in seven exons. And two non-synonymous mutations in scope of CDS(c.1615G>A and c.2938A>G), which caused the variation of amino acid. Hower,there were three SNPs loctated in3’-UTR of exon19. The results showed that most of mutation loci in the population were the moderate polymorphism and were according to Hardy-Weinberg equilibrium.In a word, the population may have a certain degree of genetic variation and selection potentials.The findings showed that there were three synonymous mutations which assciated with several carcass traits by the least-squares analysis and significance test.Then,6kinds of haplotype and12kinds of diplotype combinations were constructed. Association analysis demonstrated that diplotype H4H4was highly significantly associated with all carcass traits except thigh weight while H1H4was highly significantly associated with thigh weight(p<0.05). On the other hand, H5H6was hardly no significant effect with live weight, body weight, eviscerated weight while H3H5had no significant effect with semi-eviscerated weight, breast muscle weight and thigh weight and H2H5had no effect with abdominal fat weight. Haplotype analysis results demonstrated that H4(T-A-A) was predominant diplotypes combinations, and it may had a positive effect on promting the performance of the chicken. however its frequency was low in population that there would be a larger choice space. Therefore, the appropriate haplotype and molecular marker shoule be slectected for exploring the effective utilization of marker assisted selection in actual production and application.
