In order to study the fundus changes of vitamin A deficiency blindnessbeef cattle as well as the distribution law of associated protein. Vitamin A deficiencyin beef cattle was tested by epidemiology, behavioral, hematological, as well asfundus photography used to observe and record the fundus changes. Eye fixationmethod of the cattle was to explored. Immunohistochemical methods to explore thedistribution pattern of the protein.Body temperature of vitamin A deficiency cattle were increased, loss of appetite,blurred vision, gait changes, standing firm. Eye examination found that cow’s pupilexpansion round; incidence rate of46%.Compared with the time of normal cattle through the barrier-free access,prevalence cattle spented more time, the difference was significant. The collisionobstacle number of prevalence cattle through the barrier channel more frequently thannormal cattle, and the difference is highly significant. The through time of prevalencecattle and normal cattle with the increase of the number of repetitions, which reducedand gradually stabilized:Vitamin A content of serum and feed are far below compared with the normalstandard content. From the results of Serum biochemical parameters,we can obtainedthat the examination are normal except AST increased slightly. Blood index testresults of the prevalence cattle can be seen that MCVcontent of blood below thenormal range.The surface of disc is flat, the edge clear and visible,and the shape and size isnormal in healthy calf. The branch and color of retinal arteria and vein is basicallycommon, and No edema, hemorrhage,exudation and disorder of pigment were found.Tapetal and nontapetal fudus is normal. Ophthalmoseopically showed grosspapilloeirregular local areas in the tapetum nigrum,and more obvious in the tapetumnigrum than in the tapetum lacidum. Later they occurred in the tapetal region andappeared as small bright orange patches with a pale grey centre. The vascular tortuosity phenomenon, retinal arteries was significantly lower than the normal bovine,gray speckled injury and absorbance pigment layer appears irregular small piece ofold lesions. No other significant abnormal.Control group conventional fixed method of bovine eye can not make the retina,choroid and sclera are closely linked, retina is seriously out. While the fixed methodof bovine eye of the test group, we can found that retina, choroid and sclera areclosely linked, only a small part of the retina detachment phenomenon. The trabecularmeshwork tissue sections images of control group show that the trabecular meshworkcontinuity has been severely damaged. The test group the trabecular meshwork tissueslice image shows the trabecular meshwork continuity unspoilt. Slice images of theretinal tissue of the control group displayed severe retinal detachment and retina cellcontraction serious. The retina and choroid and sclera were separation each other.Retina tissue defect, fracture. Retinal no detachment and the organization noshrinkage of the trials group.Retinal thickness of the normal bovine and vitamin A deficiency, and the distanceof the inner plexiform layer, ganglion cell layer, the nerve fiber layer, ganglion celllayer to the inner limiting membrane, and distance of from inner nuclear layer, outernuclear layer to the outer layer, and the distance of ganglion cells to the inner nuclearlayer, and the thickness of the inner nuclear layer, outer plexiform layer, the outernuclear layer, the cone rod, the choroid. The thickness of the choroid and retina ofvitamin A deficiency cattle is less than the normal cattle,s, and difference is extremelysignificant; Compared with normal bovine, retinal layers of the prevalence bovineexhibits a distinct change. Cone and rod layer disappeared or shorter, which numbersignificantly reduced. The outer plexiform layer disappears, and the inner nuclearlayer and the outer nuclear layer overlapping. Compared with normal bovine, theblood vessels of iris and retinal and the trabecular meshwork of the prevalence bovineare significantly reduced, and iris vessel diameter aslo significantly reduced. Corneawas no significant difference.IRBP of bovine eyes organizations expression results can be drawn that theyellowish stained material existence in the inner nuclear layer, outer lamellar layer,the outer nuclear layer, the external membrane, cone and rod layer section, outersegment of cone and rod layer and the retinal pigment epithelium. There was noyellowish stained material existence in the choroid, sclera, inner nuclear layer,ganglion cell layer, nerve fiber layer, iris, cornea, inner plexiform layer. we also observed that some light yellow-stained material also existence in the choroid and irisvessels.Melanopsin of bovine eyes organizations expression can be drawn that somelight yellow-stained material existenc in the ganglion cell layer, nerve fiber layer.there was no the yellowish stained material existence in the sclera, inner nuclear layer,iris, cornea, inner plexiform layer of the inner nuclear layer, the outer lamellar layer,the outer nuclear layer, the external membrane, the inner section of the cone rod layer,layer cone rod outer segment, retinal pigment epithelium. Vascular of the choroid andiris no yellow stained material existence.We can conclude that the average gray value of IRBP and melanopsin of normalbovine eye tissue are lower than vitamin A deficiency cattle,s, and the difference wassignificant. Average grayscale value is larger, the expression amount is lower, whichcan indicate that expression amount of IRBP and melanopsin in the eye tissue ofnormal bovine are higher than vitamin A deficiency bovine, and the difference wassignificant.Conclusion: The specimens paraffin sections method of bovine eye was establishedand the distribution of IRBP and melanopsin of normal bovine; Cattle vitamin Adeficiency can cause fundus changes and eye tissue lesions and the expression of theIRBP and melanopsin are decrease.
