Duck Paramyxovirus Disease (duck-origin Newcastle disease) is an acute, highlycontagious and lethal infectious disease caused by duck paramyxovirus (duck-originNewcastle disease virus). The disease is mainly characterized by diarrhea, neurologicalsymptoms, focal glandular gastric bleeding or ulcers, intestinal mucosal bleeding and necrosisof the pancreas or the spleen. The pathogenicity of DPMV to ducks is enhanced, waterfowls(goose, duck, penguin et al.) are not only the host and reservoir of Avian Paramyxovirus Ⅰ,but also are susceptible to this virus.Avian pararnyxovirus type has been one of the mostdevastating pathogens of poultry industry,which mainly causes Newcastle disease in chicken,APMV-1infections in goose, APMV-1infections in pigeon,etc. Inrecent years, the host rangeof APMV-1has atendency to be enlarging,and it has been reported that some biologicalcharacteristics including the virulence and its associated genes, pathogenicity andimmunogenicity among the APMV-1isolates from different poultry species are not fullyidentical. Therefore, it is very necessary mainly to study the main biological characteristicsfor approach the mechanism of evolution,infection and spread of viruses, and theepidemiology of disease in the field.In recent years, the infection of Duck Paramyxovirus was on the rise, which was lead tosubstantial loss in the poultry industry. Studying on duck Paramyxo-virus has greatimportence in controlling Duck Paramyxovirus disease and protecting the development ofducking industry.Three Duck Paramyxo Viruses were isolated from shandong jinan province. TheFusion(F)gene of three isolate strains were amplified by reversetranscription-polymerasechain reaction(RT-PCR)technique.The F genes were sequenced after the products ofamplification being cloned. So that it can give great help for the study of the prevalence andcontrol.A strain virus isolated from duckling was identified as paramyxovirus by HA, HI. Thestrain was able to agglutinate chicken red blood cells, which can be inhibited by Newcastledisease virus (NDV) classic antiserum, but can’t be inhibited by avian influenza virus (AIV)classic antiserum (H5, H7, H9subtype). The virus was identificated by RT-PCR and namedSDJN01、SDJN02and SDJN03strain. LD50of duck embryos is10-7.8-10-8.2.The MDT, ICPI and IVPI for strains were50.6h-56.0and1.68-1.74respectively. The results indicated that three isolates were virulent strain.F gene of the five isolates amplified and sequence analyzed by using RT-PCR showedthat the homology between the five strains was98.5%-100%%and96.0-%~98.5%amino acidhomology to those of paramyxoviruses isolated from ducks and gooses, and homologybetween the five strains and homologies between the five strains and other ducks, goose NDVwere96.0%~98.5%. But only83.7%-84.6%nucleotide and88.6%-89.2%amino acidhomology to Lasota strain.Genetic evolution analysis showed that five strains duckparamyxovirus and Heilongjiang、Jiangsu、Fujian in the same branch.The results indicatedthat SDFCH strain was genetically closer to paramyxoviruses isolated from ducks and goosesthan to Lasota strain.
